Despite sensitivities to the wetting of crops, the overall study of the waterlogging stress is limited compared to the study of drought and other environmental stresses. The purpose of this study was to investigate the growth pattern of sesame cultivated in paddy field and the protein expression patterns of sesame leaves under waterlogging stress. All of the growth characteristics measured at the 10-leaf and flowering stage were reduced compared to the upland. Based on these results, artificial waterlogging is treated to 10-leaf stage and flowering stage for two days and three days, respectively. The leaves were collected, and proteins were separated by 2-DE and identified using LTQ-FTICR mass spectrometry. More than 300 protein spots were reproducibly detected on the CBB staining gel using Progenesis SameSpot software. Twenty-one (20) protein spots were differentially altered, and their abundance was significantly responsive to waterlogging treatment, with more than a 1.5-fold change in intensity. These differentially displayed protein spots were analyzed by mass spectrometry. A total of 11 proteins showed increased expression, and 9 proteins showed decreased expression in treated samples compared with the control seedlings. The abundance of most identified protein species, which function related to carbonization and photosynthesis is increased due to various metabolic reactions in plants due to moisture. Also, the protective mechanism against stress increased expression of a protective protein such as heat shock protein. In the case of flowering stage, more than 400 protein spots were identified using the Progenesis samespot software program. A total of 31 protein spots that exhibited more than a 1. 5-fold changes in intensity and the 31 differentially expressed proteins were successfully analyzed using mass spectrometry analysis. Among these proteins, a total of 19 proteins showed increased expression, and 12 proteins showed decreased expression in the treated samples compared to their levels in untreated plants.
