Heat treatment is one of the most effective methods for eliminating undesirable microorganisms in foods. Antibiotics are used in the treatment and prevention of bacterial infections. During food processing, food-borne bacteria encounter various environmental stress, including acid, osmotic and cold stress, and show modifications in their heat and antibiotic resistance after exposure to such food-related stresses. The main object of the present study was to evaluate the effect of food-related stresses on heat and antibiotic resistance of bacteria. In first study, the influence of food matrix on heat tolerance of residual bacteria investigated by estimating the thermal inactivation D-values (decimal reduction time) of Salmonella Enteritidis in four different liquid egg products (egg white, whole egg, egg yolk, and egg yolk + 10% NaCl). Liquid egg products inoculated with S. Enteritidis were heated in capillary tubes to temperatures ranging from 51 to 59 ℃, and then spread-plated onto tryptic soy agar, followed by incubating at 37 ℃ for 24 h. Concurrently, the performance of egg pasteurization protocols were evaluated using the calculated D-values. S. Enteritidis in egg white showed the lowest D-values followed by whole egg and egg yolk, attributing to the high pH and antimicrobial proteins of egg white. The addition of NaCl to egg yolk increased the heat resistance of S. Enteritidis. In the second study, to investigate the modifications in heat resistance and heat shock gene expression of S. Enteritidis following its prolonged exposure to combination of acid and salt stresses and subsequent cold stress, cells were grown in TSB of four pH (4.5, 5.4, 6.4, 7.3) and four NaCl concentrations (0, 1, 2 and 3%) at 37 ℃ for 24 h, and then kept at 4 ℃ for 0, 1, 4, and 7 days. For fresh (0 day) and short refrigerated (1 day) cultures, previous adaptation to low pH or high NaCl stress increased the heat resistance and gene expression of S. Enteritidis, but combination of these stresses reduced bacterial heat tolerance, specially the growth medium pH was decreased. Refrigeration period is negatively correlated (P<0.01) with the d-value (r=-0.505) and transcription levels of rpoH (r=-0.654), dnaK (r=-0.652), groEL (r=-0.645). In third study, the effect of acid, salt, and cold stress on antibiotic resistance of S. Enteritidis, Listeria monocytogenes, and Escherchia coli O157:H7 was investigated using disc diffusion methods. For S. Entertidis, acidic growth conditions increased (P<0.05) the resistance to ciprofloxacin and erythromycin, and the addition of 4% NaCl to growth mediums decreased (P<0.05) the resistance to chloramphenicol. Regardless of pH and NaCl concentration of growth mediums, refrigerated E. coli showed increased antibiotic resistance (P<0.05) to amoxycillin, ciprofloxacin, gentamicin, streptomycin, and erythromycin. Acid-adapted L. monocytogenes showed decreased antibiotic resistance (P<0.05) to ampicillin, amoxycillin, ciprofloxacin, gentamicin, streptomycin, tetracycline, erythromycin, and chloramphenicol. Present study provided an evaluation method for heat resistance environmental-stressed S. Enteritidis by comparing the D-value of S. Enteritidis. Present results demonstrated the effects of the environmental stresses on the heat resistance of S. Enteritidis. The combination of preserving methods is expected to prevent S. Enteritidis survival caused by stress adaptation or cross-protection in pasteurized food products. These results have important implications in the food industry and may facilitate more accurate risk assessments, the development of effective critical control points, and the optimization of mild thermal treatments for reducing negative effects on some foods, such as liquid egg products, dairy products, and fermented foods. Furthermore, prolonged exposure of foodborne bacteria to acid, salt, and cold stress alters their antibiotic resistance.
