QI의 투여는 전립선 비대를 억제하였고 전립선과 혈청에서Dihydrotestosterone (DHT) 와 testosterone의 수준을 유의있게 감소시켰으며 전립선에서 세포증식의 마커인Proliferating Cell Nuclear Antigen (PCNA) 와 Cycline D1의 수준을 감소시켰다. 또한 QI는 AKT 와Glycogen Synthase Kinase 3 beta (GSK-3β) 의 활성을 억제하였으며 자가사멸을 억제하는 단백질인B-cell lymphoma 2 (Bcl-2) 의 발현은 저하시키고 자가사멸을 유도하는Bcl-2-associated X protein (Bax) 의 발현은 향상시킴으로써 Bcl-2/Bax 비율을 감소시켰다. caspase-9와 -3의 발현또한 QI 투여 후 향상되었다. 이러한 결과로 미루어 보았을 때, QI는 BPH를 억제하는 효과가 있으며 BPH의 치료에 대한 잠재력이 있는 것으로 생각된다.

Quisqualis indica (QI) is a widely consumed medicinal plant to treat various health disorders in Asian countries. Plant has various biological activities including, anti-inflammatory, antipyretic, antioxidant, immunomodulatory, antistaphylococcal, and acetylcholineserase inhibitory activities. However, no reports exist on its beneficial effect against BPH. Therefore, in this study we evaluated the anti-BPH effect of QI on testosterone induced BPH in a rat model. Twenty-four Sprague-Dawley (SD) rats were divided into four groups (each n=6): control group, BPH model group, Finesteride treated group (10mg/Kg) and QI treated group (150mg/Kg). BPH was induced by 3 mg/kg subcutaneous injections of testosterone propionate for weeks. Finesteride and QI were concomitantly administrated via oral gavage. At termination, the rats were sacrificed to obtain the samples. Our results supported that the QI treatment significantly inhibited prostatic hyperplasia by improving the prostate gland morphology and reducing the DHT, testesterone level in prostates and serum of BPH induced rats. Also, QI attenuated the expression of cell proliferative markers, such as PCNA (Proliferating Cell Nuclear Antigen), Cyclin D1 in BPH induced prostates. Furthermore, our investigations showed that QI inhibited the activation of AKT and Glycogen Synthase Kinase 3 beta (GSK3β). Further, QI supressed the expression of anti-apoptotic protein, Bcl-2 and enhanced the level of pro-apoptotic, Bax and decreasing the B-cell lymphoma 2 (Bcl-2)/Bcl-2-associated X protein (Bax) ratio in prostate tissues of BPH rats. Also, we found enhanced expression of caspase-9 and -3 activities following the treatment with QI. Overall, these findings suggest that QI exert a suppressive action on BPH, thus it has a potential to be developed as a commercial drug to treat human BPH.
