In chapter 1. I studied on acid-treated water soluble chitosan microneedle(MN) for transdermal drug delivery. MN patches are minimally invasive and can be used to deliver hydrophilic therapeutics. Chitosan has been widely used as a polymeric MN material because it is abundant and biocompatible. However, it also has poor solubility and high viscosity in aqueous solutions at neutral pH limit the use of chitosan in the microneedle fabrication. Herein we report a facile way to increase the water solubility of chitosan in neutral pH (6~7.4) by a simple treatment with trifluoroacetic acid, resulting in random cleavage of glycosidic bonds in chitosan chain. This acid treatment decreased the molecular weight of chitosan and increased water-dissolving moieties such as NH2, thereby achieved 7 wt% aqueous chitosan solutions with good filling properties that are suitable for microneedle fabrication by one-step molding process. Bullet-shaped chitosan microneedle patches have been successfully prepared by solvent casting without significant shrinkage from the mold and exhibited good mechanical strength sufficient to penetrate a porcine skin. The chitosan MNs were strong enough to penetrate porcine skin, and were insoluble in wet environments, possibly as a result of physical crosslinking of the chitosan chains. Chitosan MN patches loaded with rhodamine B (a model hydrophilic drug) had zero release kinetics in tests on porcine skin, suggesting their potential for long-term transdermal delivery. The skin irritation is slight and transient, indicating the high safety of the trifluoroacetic acid(TFA)-treated chitosan MN patches. In addition, chitosan MN patch prepare from TFA-treated chitosan can also be used for applications composed to the drugs, vaccine patches, tissue adhesive, wound healing patch, cosmetics as less abrasive to the skin. The crosslinked MPs loading minoxidil, medication to treat a hair loss, shows a zero-release kinetics exhibiting 90% drug release in the course of 2 weeks and enhanced drug permeation into skin compared to conventional topical formulations. Since the crosslinking density of spray-dried MPs can be controlled by the amount of natural crosslinker (genipin), this all-natural crosslinked CS microparticle would be suitable for drug delivery carrier requiring controlled release kinetics in local or systemic drug delivery.
