Recently, therapeutic options for treating SCI have been proposed by many researchers using a variety of cell types, such as mesenchymal stem cells (MSCs) from various sources, neural stem cells (NSCs) from aborted fetal brains, as well as embryonic stem cell (ESC) or induced pluripotent stem cell (iPSC)-derived oligodendrocyte or neural precursors. However, there is no reliable treatment method for functional recovery of SCI at present. In this study, the therapeutic potentials of human umbilical cord-derived mesenchymal stem cells overexpression Olig2 (hUC-MSC.Olig2) in a rodent model of spinal cord injury were evaluated. Human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) show similar properties with other MSCs and have the advantages of easy and non-invasive procurement of cells, less ethical problems, high neurotrophic factor secretion, anti-inflammation and reduced host immune rejection reaction, compared with ESCs or iPSCs. Olig2 is a transcription factor that plays an important role in determining cell fates into motor neurons and oligodendrocytes. In order to overcome the current limitations of hUC-MSCs, including limited neurogenic and therapeutic potentials in SCI, the new hUC-MSC line that overexpresses Olig2 gene by adenoviral gene delivery was generated. Also functional improvement following transplantation into a rodent model of SCI was observed. hUC-MSC.Olig2 cells were injected intrathecally to the injury site (1.3x106cells in 10㎕) at 7 days after SCI at T11 level and observed up to 8 week using various behavior tests and electrophysiological methods. Immunohistochemical analysis indicated that the transplanted animals exhibited increased expression of neurothrophic factor (BDNF), reduced expression of pro-inflammatory markers (iNOS) and increased expression anti-inflammatory markers (CD206), compared to the control. there was the lesion cavity volume by H&E staining, apoptotic cell death by TUNEL assay and glia scar formation by GFAP expression were significantly reduced. Transplanted animals showed significant improvement of motor function, judged by BBB and CBS tests, as well as sensory function, judged by von Frey filament test. Electrophysiological analysis using MEP also exhibited that the transplanted group was functionally improved, compared with the control group. Taken together, these results strongly suggest that hUC-MSC.Olig2 is effective for treating a rodent model of SCI and can provide a therapeutic possibility for clinical application in the future.
