Background: Biomolecules are the molecules which construct organism and undertake specific function in organisms. Most of them are consisted of organic molecules with very small amount of metallic or nonmetallic elements. Certain biomolecules are used as biomarkers of disease because their expression levels are regulated by external stimuli or diseases. In order to visualize such biochemical changes at the molecular level, development of various kinds of molecular imaging probes has been continuously demanded. This thesis focuses on the development of novel fluorescent probes that selectively label target biomolecules in cells, through diversity-oriented synthesis or the conjugation with specific functional groups to fluorophores. Method: This work is divided into two tasks: the development of new fluorophores with photocrosslinking and the discovery of new probes that bind to specific biomolecules. First, photocrosslinking property makes the probe bind to adjacent biomolecules under ultraviolet light. We developed novel photocrosslinking fluorophore derived by known fluorophore. Next, we synthesized novel probes for various biomolecules. The details proceed in the order of probe development, mechanism identification, and probe application. The design of the probes utilized diversity-oriented approach or conjugation with the known functional groups. Result: First, photocrosslinker which inheres in the probes were accomplished without changing the size of the probe. Also, we firstly revealed that the simple probe which is widely used for Excited state intramolecular proton transfer (ESIPT) has photocrosslinking property. Next, we developed probes which specialized to progesterone, superoxide, palladium (II) ion and N-terminal cysteine, respectively. The mechanisms were identified through spectroscopy, mass spectrometry, and electron microscopy. Plasma from pregnant and the cells infected by avian influenza viruses was used for application. Conclusion: The results of our studies were performed to broaden the choice of tools available for molecular imaging and protein profiling in chemical biology. In particular, we discovered the fluorescent probe among steroid which have very similar structures but performs entirely different task. Also, new alternative ways to discover and analyze new probes were reported. Ultimately, we expect our research will contribute to accelerating chemical biology research.
