Autophagy is a pathway that degrades cytoplasmic materials. It formed a double membrane structure called the autophagosome which fuses with lysosome for degradation. Although autophagy has been studied for over 50 years using transmission electron microscopy(TEM), systematic analysis of autophagosome steps have not been performed. In this study, ultrastructural analysis of WT MEFs and ATG5 KO MEFs was performed. ATG5 plays an important role in elongation of the autophagic membrane. Using TEM, Firstly the ultrastructure at each step of autophagosome in WT MEFs was identified. However, the autophagosomes in ATG5 KO MEFs did not complete the membrane formation and remained immature. The number of autophagosomes was quantified based on ultrastructural analysis. These quantitative analysis method of each step of autophagosome was established by using TEM. Based on this method, we analyzed the autophagosome of Cohen syndrome and Huntington's diseasea at ultrastructural level. EM analysis revealed that the autolysosome was most abundant in fibroblasts in Cohen syndrome. These ultrastructural analysis of autophagosome suggest that autophagy dysregulation might be related to the pathogenesis of Cohen syndrome. Also, in Huntington's disease, our EM data indicates that the late autophagosome was increased in the cells expressing the mutant huntington protein. This data raise the possibility that improving the fusion step of autophagosome and lysosome in Huntington's disease, may be effective in the treatment of Huntington's disease. Take together, our study indicates that the ultrastructural and quantitative analysis of autophagosome steps using TEM can be applied to various human disease models related to autophagy.
