CHAPTER 2: 제2형 당뇨병 모델로 이용되고 있는 db/db mouse에서 양춘사 물 추출물의 항당뇨 효과를 조사하였다. 양춘사 추출물을 100, 200, 500 mg/kg의 농도로 4주간 경구 투여한 결과 공복시 혈당을 유의하게 억제 하였으며, 또한 경구 당부하 검사에서도 유의한 혈당 상승 억제 효과를 나타냈다. 양춘사 추출물의 항당뇨 효과 기전을 조사하기 위하여 간 조직에서 지방합성과 관련된 유전자(FAS, ACC, SREBP1)의 발현 정도와 염증과 관련된 인자들(iNOS, COX, NF-κB)을 조사하였다. 그 결과 양춘사 추출물을 투여한 군에서 FAS, ACC, SREBP1 mRNA 발현이 대조군(양춘사 추출물을 투여여하지 않은 db/db mouse)에 비해 유의하게 억제되었다. 또한 간에서 염증 유발 인자인 NO와 PGE2의 생성과 관련된 iNOS, COX-2 단백질 발현이 양춘사 추출물에 의해 유의하게 억제되었다. 이러한 효과는 iNOS, COX-2의 발현을 조절하는 전사인자인 NF-κB의 활성화 억제에 기인한 것으로 조사되었다. 이러한 결과는 양춘사 추출물이 제2형 당뇨병 치료제 개발의 후보 물질이 될 수 있음을 시사한다. Methods: This study examined the in vitro inhibitory effect against rat small α-glucosidase and porcine pancreatic amylase by using AVE of different concentrations. In addition, after oral administration of AVE to rats, the in vivo sucrose and amylose lading test was carried out to measure blood glucose concentration levels.
Results: The results of the in vitro experiment show that AVE had an inhibitory effect against α-glucosidase in the positive control group like acarbose but did not have an inhibitory effect against α-amylase unlike acarbose. AVE showed a significant inhibitory effect against postprandial blood glucose in the in vivo sucrose loading test but showed little inhibitory effect in the amylose loading test.
Conclusions: It is believed that the results of this study can provide basic data for the development of type 2 diabetes treatments that have the same medicinal effect as acarbose, α -glucosidase inhibitor, but that can reduce side effects resulting from the use of acarbose.
