G0S2가 침습성 MDA-MB-231 유방암 세포의 악성 표현형에 영향을 주는 지 확인하기 위해서, G0S2 siRNA를 사용하여 G0S2의 발현을 억제하였고, G0S2의 억제가 세포 생존 능력, 증식, 이동 및 침윤을 감소시키는 지 관찰하였다. 또한, 상세한 분자 기전을 규명하기 위해, 발암 신호 경로와 G0S2의 상관 관계를 탐구하였다. 결과적으로, G0S2의 발현을 저해했을 때, integrins 조절에 의한 FAK-Src 신호전달이 불활성화 되어Hippo pathway 신호전달이 활성화되는 것을 확인하였고, ERK1/2 신호 전달은 비활성화 되는 것을 확인하였다. 또한, G0S2 억제가 β-catenin의 단백질 발현 수준을 감소시키는 반면, E-cadherin의 단백질 발현을 증가시킨다는 것을 확인하였다. 결과적으로, G0S2는 Hippo pathway를 조절하고, EMT유도하여 MDA-MB-231 유방암 세포의 생존과 전이에 기여하는 주요 인자일 수 있음을 최초로 밝혔다.

Human breast cancer cell line MDA-MB-231cells are highly invasive and aggressive, compared to less invasive MCF-7 breast cancer cells. To identify the genes that might be related to the malignant characteristics of MDA-MB-231 cells, microarray analysis was performed in MDA-MB-231 and MCF-7 cells. The result showed that the level of elevation in the expression of G0S2 was highest among upregulated genes in MDA-MB-231 cells. The most well-known function of G0S2 is the direct inhibition of adipose triglyceride lipase (ATGL). However, the function of G0S2 in cancer progression is not yet understood. To investigate whether G0S2 affects phenotypes of invasive MDA-MB-231 cancer cells, G0S2 expression was suppressed using G0S2 siRNA and observed that inhibition of G0S2 decreased cell viability, proliferation, migration, and invasion in MDA-MB-231 cells. To explore its detailed molecular mechanisms, the correlation of G0S2 with oncogenic signaling pathways was investigated. Consequently, it was found that inhibition of G0S2 inactivates integrin-regulated FAK-Src signaling which allows the Hippo signaling to be active, and it also inactivates ERK1/2 signaling. In addition, suppression of G0S2 decreased the protein expression level of β-catenin while the expression level of E-cadherin was increased. My results show for the first time that G0S2 mediates the Hippo pathway and induces EMT. Taken together, this study suggest that G0S2 could be a major factor contributing to cell survival and metastasis of MDA-MB-231 breast cancer cells.
