인플루엔자는 바이러스성 호흡기 감염 질환 중 하나이며 인플루엔자 A 바이러스는 조류, 돼지, 말, 해양 포유 동물 및 인간을 비롯한 다양한 종을 감염시킬 수 있다. 그 중 야생 철새는 조류 인플루엔자 바이러스 (AIV)의 알려진 자연저장소로 저병원성 인플루엔자 바이러스가 야생 조류에서 가금류로 전염 될 때 재조합 혹은 변이를 통해서 고병원성 인플루엔자 바이러스를 유발 할 수 있으며 그 결과 많은 경제적 손실을 초래하며 사람에게도 감염되어 인명피해를 입히기 때문에 조류 인플루엔자 바이러스 감시를 통해 새로운 유형의 바이러스를 미리 검출하여 분석하는 것은 필수적 이다.
조류 인플루엔자 바이러스 감시를 위해 2016-2017 년 동절기에 대한민국 주요 야생 철새 도래지에서 2,184 개의 조류 분변 샘플을 수집 하였고, 24 개 (1.1%)의 조류 인플루엔자 바이러스를 분리 했다. 분리된 바이러스는 전체 서열 분석을 통한 계통 발생 분석 결과 아시안 또는 유라시안 계통으로 확인 되었으며, 쥐를 이용한 병원성 실험 결과 4 개의 바이러스가(H1, H6 아형) 쥐에게 효과적으로 감염되어 높은 치사율을 보이는 것으로 확인되었다. 병원성을 보이는 바이러스들은 병원성 인자를 찾기 위해 8 개의 분절 게놈의 서열과 이전에 병원성을 증가시키는 것으로 보고된 아미노산 잔기들을 스크리닝 하였고, 그 결과 PA 그리고 PB1-F2 프로테인에서 병원성을 증가시키는 아미노산 돌연변이를 확인 했다 (PA 프로테인 383 번 잔기의 아스파르트산 치환, PB1-F2 프로테인 62 번 잔기의 류신 치환, 73 번 잔기의 리신 치환, 79 번 잔기의 아르기닌 치환, 82 번 잔기의 류신 치환 그리고 PB2 프로테인 504 번 잔기의 발린 치환). 전체 분변 샘플 중 무작위로 202 개(9.2%)에 대한 새 종 분석 결과 모두 기러기 목에 속했으며 분석된 샘플 중 43.5% 그리고 인플루엔자 양성 분변 샘플 중 85%가 회색기러기 종에 속했다. 이 결과로 보아 회색기러기 종이 2016-2017 년 겨울철에 한국으로 들어온 가장 흔한 종이며 조류 인플루엔자 바이러스의 주요 운반체라고 생각된다.

Influenza is one of the most common causes of viral respiratory infection, and it can be divided into types A, B, C and D. Influenza A viruses can infect a variety of species including birds, swine, horses, marine mammals and humans. As such, waterfowls are known carriers of avian influenza virus (AIV). Transmission of AIV from wild-birds to poultry can cause the highly pathogenic avian influenza (HPAI) in poultry, resulting in economic loss. Furthermore, some AIVs can cause zoonotic disease and pandemics. Therefore, AIV surveillance in wild birds is essential for animal and public health. In this study, we collected avian fecal droppings from major wild bird habitats in South Korea during the 2016-2017 winter season for AIV surveillance. A total of 2,184 fecal samples were collected, of which 24 samples (around 1.1%) were revealed to be AIV-positive. The AIV were isolated from the positive samples, and each isolate was analyzed thru full-genome sequencing, phylogeny and mouse pathogenesis. We also identified the bird species of origin of 202 random fecal samples (around 9.2%). Our analyses showed that the isolates represented six HA subtypes (H1, H3, H5, H6, H7 and H8), and they can be divided into two lineages: Asian and Eurasian. Four isolates (#6173-H1, #6535-H6, #6536-H6 and #7134-H6) were able to efficiently infect mice; however, further studies about the cause of high virulence in mice will be required. Based on the analyzed data, eight bird species have been identified, all in the order Anseriformes, and around 85% of the AVI-positive samples were carried by the species A.anser. These results suggested that the greylag goose (A. anser) was the most prevalent species that migrated to South Korea during the winter season of 2016-2017 and was the major carrier of AIV.
