It is well-known that the fusion between a vesicle and a target organelle is mediated by the formation of SNARE (soluble N-ethylmaleimide-sensitive factor adaptor protein receptor) complex. Although the syt4 syt5 plants retain immune responses to the Erysiphe pisi fungus, they show compromised immune responses to surface-inoculated Pseudomonas syringae DC3000 bacterium. SYT5 directly interacts with all SYP132, SNAP33 and VAMP722 SNARE proteins, which is promoted by Ca2+. In addition, we also found that binary interactions between SYP132, SNAP33 and VAMP722 are Ca2+-dependently facilitated by SYT5. The impaired resistance to surface-inoculated P. syringae DC3000 in VAMP721/722-depleted plants suggests that SYT4/5 regulated immune responses to bacterial pathogens by Ca2+-dependently promoting the formation of SYP132-SNAP33-VAMP721/722 SNARE complex. ER stress triggered by diverse external stimuli was reported to inhibit plant growth, in which CRT1/2 (calreticulin 1/2) are critically important. We found that VAMP721/722 expression was increased by ER stress inducers such as TM (tunicamycin), DTT (dithiothreitol) and AZC (L-azetidine-2-carboxylic acid). Interestingly, VAMP721/722 protein levels were no more elevated in the crt1/2 plants by TM, although TM still increased VAMP721/722 transcription in those plants. Re-elevation of VAMP721/722 protein levels by TM in MG132-pre-treated crt1/2 plants, and in vitro and in planta interactions between CRT1/2 and VAMP721/722 suggest that CRT1/2 play an important role in stabilizing VAMP721/722 proteins during ER stress responses. Compared to WT plants, growth in VAMP721/722-depleted plants was more inhibited by TM.
