본 연구는 알츠하이머를 유발한 흰쥐에게 유산소 운동 방법 중 하나인 트레드밀 훈련을 강도에 따라 적용하였다. 운동의 강도에 따른 인지기능, 운동기능과 신경영양성인자인 IGF-1, NGF 그리고 BDNF에 미치는 영향을 분석하였다. 알츠하이머가 유발된 흰쥐를 저강도 운동군(Group Ⅰ, n=8), 중강도 운동군(Group Ⅱ, n=8), 고강도 운동군(Group Ⅲ, n=8), 대조군(CG, n=8)으로 무작위 분류하였다. 대조군을 제외한 각 군은 20분씩 주 5회, 총 4주 동안 운동을 실시하였다. 인지기능 평가를 위해서 radial 8-arm maze test를 실시하였고, 운동기능을 평가하기 위해 beam-walking test와 ladder rung walking test를 실시하였고 신경영양성인자 평가를 위해 insulin-like growth factor-1(IGF-1), nerve growth factor(NGF) 그리고 brain-derived neurotrophic factor(BDNF) 수치를 측정하였다. 실험결과 radial 8-arm maze test에서 그룹 간 8개의 먹이를 정확하게 찾아내는 데 걸린 시간, 동일한 먹이를 찾은 오류의 횟수와 첫 번째 오류가 발생하기 전 정확히 먹이를 찾은 횟수에서 유산소 운동을 일체 하지 않은 대조군(CG)에 비해 저강도 운동군(Group Ⅰ), 중강도 운동군(Group Ⅱ), 고강도 운동군(Group Ⅲ)에서 모두 통계학적으로 유의한 차이를 보였다. 동일한 먹이를 찾은 오류의 횟수에서는 대조군을 제외한 모든 군에서 운동 전과 후에 차이를 보였으며, 첫 번째 오류가 발생하기 전 정확히 먹이를 찾은 횟수에서는 모든 군에서 운동 전과 후에 차이를 보였다. 그리고 그룹 내 8개의 먹이를 정확하게 찾아내는 데 걸린 시간에서는 고강도 운동군(Group Ⅲ)에서만 운동 전과 후에서 유의한 차이를 보였다. 운동기능 평가에서는 두 검사 모두 그룹 간 비교에서는 대조군(CG)에 비해서 저강도 운동군(Group Ⅰ), 중강도 운동군(Group Ⅱ)과 고강도 운동군(Group Ⅲ) 모두 유의한 차이를 보였지만 운동 강도에 따른 유의한 차이는 나타나지 않았다. 그리고 그룹 내 운동의 효과 비교에서는 운동을 적용한 그룹 모두 운동 전에 비해 운동 후에 유의한 증가를 보였다. 신경영양성인자 측정에서 IGF-1와 NGF 결과는 대조군(CG)에 비해서 저강도 운동군(Group Ⅰ), 중강도 운동군(Group Ⅱ)과 고강도 운동군(Group Ⅲ) 모두 유의한 차이를 보였지만 운동 강도에 따른 유의한 차이는 나타나지 않았다. 그리고 BDNF에서는 대조군(CG)에 비해서 저강도 운동군(Group Ⅰ), 중강도 운동군(Group Ⅱ)과 고강도 운동군(Group Ⅲ) 모두 유의한 차이를 보였고, 고강도 운동군(Group Ⅲ)이 저강도 운동군(Group Ⅰ)과 중강도 운동군(Group Ⅱ)에 비해 월등하게 BDNF 발현량의 수치가 높아졌다. 본 연구의 결과를 종합해보면, 유산소 운동은 알츠하이머병 흰쥐의 인지기능, 운동기능과 신경영양성인자 발현을 향상시키는 것으로 나타났다. 그 중 고강도 유산소 운동의 경우 인지기능과 신경영양성인자의 변화에 보다 큰 효과를 나타냈다. 이러한 결과들은 트레드밀 운동이 알츠하이머병 환자의 효과적인 치료방법에 있어 유용한 기초자료가 될 것이라 생각된다.

This study applied a treadmill training as a method of aerobic exercise to rats in order to analyze effects of varied exercise intensities on cognitive function, motor function, and neurotrophic factors including. The Alzheimer's induced rats were randomly divided into a control group(CG, n=8), a group of low-intensity exercise(Group Ⅰ, n=8), a group of mid-intensity exercise(Group Ⅱ, n=8), and a group of high-intensity exercise(Group Ⅲ, n=8). The group I, Ⅱ, and Ⅲ underwent the treadmill exercise for 20 minutes, five times week, for four weeks. A radial 8-arm maze test was performed for assessing cognitive functions. A beam-walking test and a ladder rung walking test were performed for determining neurological motor functions. The values of IGF-1, NGF, and BDNF were measured for assessing neurotrophic factors. The results of the study are as follows. According to the radial 8-arm maze test, Group Ⅰ, Group Ⅱ, and Group Ⅲ showed statistically significant differences in comparison to the CG in the time taken to correctly find eight feeds between groups, the number of errors in finding the same feed, and the number of correctly finding a feed before the first error. In the number of errors in finding the same feed, all the groups except for the control group showed differences before and after the intervention. Meanwhile, in the number of correctly finding a feed before the first error, all the groups showed differences before and after the intervention. In the time taken to correctly find eight feeds within group, only Group III showed significant difference before and after the intervention. In both tests for motor functions, the three exercise groups, when compared to the control group, showed significant differences between groups, but without significant differences based on exercise intensity. When the effects within group were compared, the three exercise groups showed significant increases after the intervention rather than before the intervention. When neurotrophic factors were measured, showed significant differences in Group Ⅰ, Group Ⅱ, and Group Ⅲ when compared to that of the control group, but without significant differences based on exercise intensity. BDNF showed significant differences in Group Ⅰ, Group Ⅱ, and Group Ⅲ when compared to that of the control group, and the expression ratio was markedly higher in Group Ⅲ than in Group Ⅰ and Ⅱ. In conclusion, aerobic exercise can improve cognitive functions, neurological motor functions, and expression of neurotrophic factors in Alzheimer's induced rats. In addition, high-intensity aerobic exercise, in particular, can induce more effects on changes in cognitive functions and neurotrophic factors. The results of this study can be provided as useful basic data in developing treatments for Alzheimer's disease patients.
