Ty-2 는 최근 Nucleotide-binding domain and leucine-rich repeat-containg (NB-LRR) 를 암호화하고 있는 것으로 보고되었다. 저항성과 이병성의 염기서열 상의 차이를 비교하였고 15개 SNP를 확인하였으며, 그 중 1개의 SNP를 바탕으로 cleaved amplified polymorphic sequence (CAPS) 분자표지로 제작하였다. Ty-4 는 암호화하고 있는 유전자는 밝혀지지 않았으나 염색체의 550 kb 구역 내에 65개 유전자가 밝혀져 있다. 그 중 Serin/threonine-protein phosphatase 유전자와 Myb-like protein 유전자에서 저항성과 이병성 염기서열의 차이를 비교하였고 Serin/threonine-protein phosphatase 유전자에서는 Insertion/deletion (InDel) 분자 표지, Myb-like protein 유전자에서는 derived-CAPS (dCAPS) 분자표지를 제작하였다. 본 연구에서 개발된 TYLCV 저항성 분자표지는 병 저항성 육종의 효율과 정확도 향상에 크게 기여할 것으로 기대된다. 다양한 TYLCV 유전자원에 TYLCV 저항성 분자표지를 적용한 결과와 함께 TYLCV 저항성 유전자원의 저항성 정도를 분석하기 위해 생물 검정(bioassay)를 통해 표현형과 유전형 평가를 함께 실시하였다. 생물 검정 결과를 종합해보았을 때 TYLCV 저항성을 한 개 가지고 있는 단일 저항성 보다 두 개 이상 가지고 있는 복합 저항성에서 높은 수준의 저항성을 보였으며 단일 저항성 계통은 비교적 약한 저항성을 확인할 수 있었다. 그리고 국내 육종회사에서 판매되고 있는 39개 시판품종에 TYLCV 저항성 분자표지를 적용해본 결과 육종회사에서 제공한 TYLCV 저항성은 대부분 Ty-1/3 저항성인 것으로 확인되었다. 생물 검정 결과와 같이 단일 저항성보다 Ty-1, Ty-2, Ty-3, Ty-4, ty-5, Ty-6 피라미딩을 통해 복합 저항성 품종을 육성하여 높은 수준의 TYLCV 저항성 품종을 육성하는 것이 필요하다.

Among several TYLCV resistance genes known to date, Ty1 resistance has been most intensively investigated and utilized in the TYLCV resistance breeding programs. Recently developed TYLCV resistance varieties with Ty1 gene are on the market, however, exploration of novel resistance sources and resistance gene pyramiding is still to achieve durable and stable resistance against TYLCV. TYLCV resistance genes reported to date include Ty1/3, Ty2, Ty4, ty5 and Ty6 derived from wild-type tomatoes. Ty1 and Ty3 located on chromosome 6 are found to be allelic and originated from S. chilense and encodes an RNA-dependent RNA polymerase (RDR). Ty2 derived from S. habrochaites is located on chromosome 11, encodes an nucleotide-binding domain and leucine-rich repeat containing (NB-LRR) gene. Ty4 resistance was reported in S. chilense line LA1932 in addition to Ty3 and shown to increase the level of the resistance. A recessive resistance gene ty5 derived from S. peruvianum is located on chromosome 4 and loss-of-function mutant allele of the pelota gene. Another recessive resistance gene Ty6 derived from LA1938 and LA2779 was recently mapped to chromosome 10. In addition to the development of series of molecular markers on these TYLCV resistances, we focused on the development of molecular markers for the Ty2, Ty4 resistance. Ty2 exists NB-LRR gene with using sequence information from numerous SNPs and InDels detected in these regions, we developed several InDel, CAPS markers. Ty4 exists within the 550 kb regions on chromosome 3 including approximately 30 genes. We have focused on 8 genes located in this regions, and sequence analysis of these genes was performed. Using sequence information from numerous SNPs and InDels detected in these regions, we developed several InDel, CAPS markers. In order to obtain more durable and higher levels of resistance, we are in the process of combine these resistances in multiple combinations. Infectivity assays using infectious TYLCV clones are performed to precisely address the speed and strength of the infectivity in each combination of the resistance genes.
