C. neoformans var. neoformans, C. neoformans var. grubii와 대치배양한 결과 갈변 반응이 나타나지 않았다. 따라서 표고버섯 균주에 나타나는 갈변 현상과 생장 저해는 C. pseudolongus와 표고버섯 균주간의 특이적인 관계에서 나타나는 현상으로 사료되었다. C. pseudolongus CBS8297과 DUCC4014의 효소 분비 특성을 확인하기 위해서 API 20C AUX kit와 7 가지 세포외효소 활성에 대하여 7 가지의 단일 탄소원 기질이 각각 들어간 Congo red 발색배지를 사용하여 확인하였다. API 20C AUX kit를 통해 확인하였을 때, C. pseudolongus가 사용 가능한 기질은 20 가지 중에서 17 가지로서 인체병원성 진균인 C. neoformans의 기질 이용 특성과 큰 차이를 보였다. 반응이 확인된 기질 종류 또한 다른 Cryptococcus spp.와 상당히 다르게 나타났다. 세포외효소 활성은 C. pseudolongus CBS8297과 DUCC4014 모두 protease 활성만 약하게 나타났고 amylase, avicelase, β-glucosidase, carboxymethyl cellulase, pectinase, xylanase에 대해서는 활성을 나타내지 않았다. C. pseudolongus CBS8297과 DUCC4014의 ITS rDNA, 28S rDNA, 18S rDNA, β-tubulin 유전자, tef1-α 유전자 부위의 염기서열 분석 후 유사도를 비교하였을 때, ITS와 tef1-α 유전자에서 약간의 염기서열 차이가 있었지만 다른 3 가지 유전자 부위에 대해서는 100% 서열이 일치하였다. C. pseudolongus DUCC4014의 유전체를 분석을 한 결과 유전체의 길이는 25 Mbp, 유전자의 개수는 10,257개로 추정되었다. 추정된 유전자 중에는 표고버섯의 균사체 세포벽에 영향을 줄 가능성이 있는 chitinase, glucanase, lyase 등을 코딩하는 유전자와 산화 환원 반응으로 갈변 반응에 영향을 줄 것으로 예측되는 oxidase, peroxidase, reductase를 코딩하는 유전자도 존재하였다. 본 연구 결과를 종합해보면 해외 균주인 C. pseudolongus CBS8297과 국내 균주인 C. pseudolongus DUCC4014 간의 생육 특성, 병리 특성, 효소분비 특성은 유의미한 차이를 보이지 않았다. 향후 본 연구 결과는 C. pseudolongus에 대한 병해에 대한 심층적 연구를 위한 생리학적, 유전학적 기초 자료로 활용될 것으로 기대된다.

In 2014, brown rot disease caused by the yeast Cryptococcus pseudolongus has emerged as a new problem on oak mushroom fruit bodies. However, there is little known information about the yeast. Therefore, this study was conducted to investigate the physiological and genetic characteristics of C. pseudolongus. For the investigation, C. pseudolongus DUCC4014 isolated in 2014 and the type strain of C. pseudolongus CBS8297 were used including human pathogenic strains of C. neoformans var. neoformans and C. neoformans var. grubii as the comparative species. As the hosts of these Cryptococcus strains, strains of oak mushroom cultivars, Sanjo 701ho, Seolbackhyang, Chamahram, and L808, were used. No difference in physiological characteristics was found between C. pseudolongus CBS8297 and DUCC4014. Browning induction and growth inhibition against all the mushroom strains was also found to be species-specific features of C. pseudolongus. When C. pseudolongus CBS8297 and DUCC4014 were grown in different media for three days, they initiated differentiation on CMA and MEA from yeast form to pseudohyphae form. When these were cultivated for 7 days, pseudohyphae were also observed on the other media such as CYA, MEA(Blakeslee'), PDA, V8-juice agar, Winge agar, and YMA. In dual culture assays, PD broth cultured cells of C. pseudolongus did induce browning and growth inhibition of L. edodes, but its culture filtrate did not. The cells of C. neoformans var. neoformans and C. neoformans var. grubii also could not induce browning and growth inhibition of L. edodes. The browning and growth inhibition are assumed to be species-specific phenomenon between L. edodes and C. pseudolongus. To understand the enzyme secretion feature of C. pseudolongus CBS8297 and DUCC4014, the API 20C AUX kit and Congo red chromogenic media containing each of seven different substrates were tested. With 20 substrates in API 20C AUX kit, C. pseudolongus was able to use 17 substrates of them. The number of substrate usage differed between C. pseudolongus and a human pathogenic fungus C. neoformans. The intensity of substrate degrading activity also differed between C. pseudolongus and other Cryptococcus spp. tested. On the chromogenic media, C. pseudolongus CBS8297 and DUCC4014 showed only protease activity weakly. No amylase, avicelase, β-glucosidase, carboxymethyl cellulase, pectinase and xylanase were detected. When the nucleotide sequences of ITS rDNA, 28S rDNA, 18S rDNA, β-tubulin gene, and tef1-α gene were compared between C. pseudolongus CBS8297 and DUCC4014, slight differences were found only in ITS rDNA and tef1-α gene sequences. But they showed 100% sequence homology in other regions. The whole genome sequencing of C. pseudolongus DUCC4014 revealed that its genome size is 25 Mbp and the number of predicted genes is 10,257. Among these predicted genes, the genes coding of chitinases, glucanases, and lyases, which may affect the cell wall of mushroom, and oxidase, peroxidase, and reductase, which are assumed to be involved in mycelial browning through oxidation-reduction reaction, were predicted by a bioinformatic tool. This study confirmed that there is no significant difference in the growth, pathological behavior, and enzyme secretion characteristics between the foreign strain C. pseudolongus CBS8297 and the domestic strain C. pseudolongus DUCC4014. The results of this study are expected to be used as basic physiological and genetic data for in depth study on the pathogenesis of C. pseudolongus.
