Therefore, the use of plant growth-promoting bacteria (PGPB) in an effort to reduce stress caused by salt is emerging as a cost-effective and environment-friendly method. In this study, the purpose was to isolate the salt-tolerant bacteria from the rhizosphere soil and identify their ability to promote plant growth under salt stress condition. Among 19 colonies isolated from rhizosphere soil, the strains screened for plant growth promotion were selected and identified by a 16S rRNA gene sequence analysis. The isolates that showed plant growth-promoting activity were strains KST-1, KST-2, AST-3, and AST-4. The closest relatives were Bacillus cereus to KST-1 and KST-2, and Bacillus thuringiensis to AST-3 and AST-4. All the selected isolates were able to survive in a high concentration of salt environment up to 7% NaCl, and 4% NaCl did not significantly affect microbial growth. It was also confirmed, when grown with crop, the strains were able to produce the indole acetic acid, but the concentrations were relatively lower than the previously reported some of the PGP bacteria. As a result of the growth experiment, all the four strains had only minor effect on the length of the barley. However, the concentration of chlorophyll in the barley leaves were found to be higher by inoculation of 4 strains than the chlorophyll concentration in the uninoculated control. In particular, the chlorophyll concentration in the Bacillus cereus strain AST-4 was 5.45 times higher than that of the uninoculated control under the same experimental condition. The isolates, B. cereus KST-1, B. cereus KST-2, B. thuringiensis AST-3, and B. cereus AST-4 were tolerant to 7% of NaCl and the wrer found to promote growth of barley in saline soil by increasing the chlorophyll content of barley leaves in salt stress environment.
