Research on human neurological diseases and brain development is limited by the lack of advanced in vitro models that are able to recapitulate the complexity of the human brain. In this regard, the organoid technology has been developed as a novel platform for modeling various diseases in vitro as well as for understanding organogenesis in vivo by closely recapitulating the in vivo pathophysiology. Recent studies have described the generation of 3D midbrain-like organoids (MOs) containing midbrain-specific cell types including functional dopaminergic (DA) neurons and neuromelanin. MOs have the potential to be used for studying Parkinson’s disease which represents the neuro-degeneration of the midbrain. However, MOs derived from human embryonic stem cells (hESCs) in previous study have deficiency for disease modeling since they couldn’t represent patient-specific disease mechanism. Here, we first generated human induced pluripotent stem cells (hiPSCs) from human neonatal fibroblasts as an alternative source of hESCs and then generated MOs under protocol used in previous study. Generally, MOs derived from hiPSCs stably grow during long term culture and keep healthy condition. MOs contain not only midbrain dopaminergic neurons but also other neuronal subtypes as well as functional glial progenitors. Furthermore, MOs are midbrain specific, while no other regional identities such as forebrain and hindbrain are exhibited. Overall, this study suggests that our hiPSCs derived MOs may be an advanced in vitro model for studying midbrain and its related diseases.
