In this study, the disease symptoms that look very similar to the symptoms of anthracnose were observed in walnut trees of Andong, Yecheon, and Yeongyang in June 2016. The lesions on the leaves initially appeared as brown spots and formed yellow halo around them, and in the case of fruit, the area around the lesions turned brown, and later rotted, followed by oozing out the bacterial leakage from the infected region. Pathogens were isolated from the disease infected leaves and fruits of walnut and cultured on to yeast dextrose carbonate (YDC) agar plates and bacterial colonies appeared in yellow. The isolated bacteria were identified using physiological and biochemical diagnostic methods such as Gram staining, Bergey's manual, and 16S rDNA sequencing. In addition, PCR amplification using a specific primer was carried out to identify pathovar as Xanthomonas arboricola pv. juglandis (X.a.pv. juglandis). Its morphological characteristics were investigated by transmission electron microscopy (TEM) and identified by BiOLOG system and was finally confirmed as X. a. pv. juglandis. In order to investigate the molecular characteristics, the multi locus sequence typing method were analyzed in comparison with the overseas strains. Andong strains were grouped with European strains and nutrient strains were grouped with Chinese strains. Based on Koch's hypothesis, isolated pathogenicity assays were performed. To confirm pathogenicity, the leaves, fruits and branches of 3-year-old walnut plants were inoculated with bacterial pathogen suspensions as a foliar spray. Control plants were sprayed with sterile distilled water (SDW). After inoculation, the gray spots on the leaves and yellow halos around the spots were developed. Fruits showed brownish rotting, and stem showed brownish symptoms, and similar symptoms appeared under field conditions. The pathogen X. a. pv. juglandis was re-isolated from all symptomatic tissues to fulfill the Koch’s postulates and was determined to be morphologically identical to the original isolates. On the other hand, since the symptoms were very similar to anthracnose caused by Colletotrichum gloeosporioides, the disease progression was correlated. When the walnut fruits were inoculated with combined pathogens X. a. pv. juglandis and C. gloeosporioides, the disease symptoms were greater in comparison with when inoculated alone. The in vitro antibiotic test revealed that X. a. pv. juglandis was sensitive to the antibiotics such as Oxytetracycline, Streptomycin, Copper Sulfate in a sequencing manner as per the increasing the zone of inhibition. This study is the first to report a walnut Bacterial blight disease that has been registered as a quarantine management disease in Korea, and a control system should be established using antibiotics that have been susceptible to disease.
