While chloroplasts accumulate at the cell surface to utilize light efficiently under weak light conditions, they move to the cell edge to avoid photodamage under strong light conditions. The chloroplast movement is mediated by rearrangements of chloroplast actin (cp-actin) filaments according to the intensity and position of light that is sensed by the blue light receptor, phototropin. CHLOROPLAST UNUSUAL POSITIONING 1 (CHUP1) is essential for chloroplast movement as the cp-actin nucleation factor. The CHUP1 C-terminus actin polymerization domain (CCAP) has structural similarity to formin homology 2 (FH2). In this study, we identified four CHUP1 C-TERMINAL HOMOLOG genes (CHUC1~CHUC4) in Arabidopsis genome. The CHUC1~CHUC4 have high sequence homology in the proline rich (FH1 like) and CCAP-like domains with CHUP1. CHUC promoted actin polymerization in the presence of profilin as the CCAP domain. However, CHUC was not involved in chloroplast movement indicating that CHUC1~CHUC4 have diverse physiological roles in Arabidopsis. Consistently, the four CHUC genes exhibited the tissue-specific expression patterns. CHUC1 and CHUC2 were evenly expressed in all tissues, CHUC3 was specifically expressed in stem, and CHUC4 was specifically expressed in pollen. In addition, chup1 and chuc1, chuc3 mutant plants showed slower gravitropic response than wild type plants, indicating that CHUC1 and CHUC3 function in gravitropic response.
