Specimens were collected in Seoul, Buan, Jindo and Busan. All the collected individuals extracted DNA, amplified COI genes of the mitochondria to determine the species. The mitochondria DNA (CYTB, 16S rRNA) and nuclear DNA (28S rRNA) genes were amplified and sequencing was performed. Genetic diversity, Haplotype network analysis, pairwise FST analysis, Analysis of Molecular Variance (AMOVA), and phylogenetic analysis were conducted based on the sequence. COI showed seven haplotypes, CYTB represented five haplotypes, 16S rRNA represented three haplotypes, and 28S rRNA represented one haplotype. Using COI genes with the most diverse haplotypes, the phylogenetic tree was analyzed based on individuals of other country from GenBank and collected in this study. Meanwhile, using NGS (next-generation sequence analysis method) through Illumina Nextseq platform, explored ten markers and developed them. Genetic diversity, pairwise FST, Analysis of Molecular Variance (AMOVA) and Bayesian cluster analysis were conducted using the ten markers developed. It is assumed to have a high gene flow within the group, and the ability of Lucilia sericata to move long distances and to propagate themselves over the effects of previously known genetic movements and geographical distances.
