N-설포닐 알디민은 일반적으로 수분에 민감한 축합 반응을 통해 1차 아민과 알데하이드로부터 합성할 수 있다. 그러나, 같은 방법으로 케톤을 사용하는 경우에는 상대적으로 반응성이 많이 떨어지기 때문에 N-설포닐 케티민을 합성하기 어렵다. 더욱이, N-설포닐 케티민을 합성하는 방법은 많이 알려져 있지 않기 때문에 새로운 합성법을 찾아내는 것은 중요하다. 실릴-케티민을 역 극성 이민으로 만들고, 친핵체로서 교차 결합 반응에 참여한다. 반응을 위해 다양한 팔라듐 촉매, 리간드, 용매 및 첨가제를 사용하여 반응 조건을 최적화하였다. 최적의 반응 조건에서, 대칭 및 비대칭 디아릴요오도늄 염과 실릴-케티민을 팔라듐 촉매 반응을 통해 다양한 디아릴케티민 화합물을 합성하였다.

N-sulfonyl imines are used in various organic reactions and can be synthesized via several reported methods. N-sulfonyl aldimine is generally prepared from a primary amine and an aldehyde by the condensation using a Lewis acid. However, N-sulfonyl ketimine is relatively hard to synthesis through the same reaction condition due to low reactivity. Moreover, discovery of new synthetic method is important since little is known to synthesize N-sulfonyl ketimines. Silyl-ketimines serve as an imine anion equivalents via reversed-polarity, and participate in coupling reactions as a nucleophile. The coupling reactions were optimized using various palladium catalysts, ligands, solvents and additives. With the best reaction conditions, various N-sulfonyl ketimines were synthesized from palladium catalyzed cross-coupling of diaryliodonium salts and silyl-ketimines.

It has been identified that activating mutations of SHP2 is related to various diseases, such as Noonan syndrome and various cancers, including leukemia, lung and breast cancer. Therefore, the discovery of SHP2 inhibitors can be an important treat of cancers. High-throughput screening (HTS) was performed using more than 10,000 compounds from Korea Chemical Bank and a hit compound with a 0.097 μM of IC50 value was obtained. We designed and synthesized various derivatives of the hit compound containing benzotropolone core to explore the structure-activity relationship (SAR) study. Biological activity was also studied in a MDA-MB-468 cell line associated with a breast cancer.
