The hyperthermophilic archaeon Thermococcus onnurineus NA1 could grow on formate and produce H¬2. Formate conversion to H2 in the strain was mediated by a formate-hydrogen lyase (FHL) complex, and this strain has been reported to grow on formate coupled with H¬2 production and ATP generation (HCOO- + H2O→HCO3- + H2, ΔG80oC = -2.6 kJ/mol). In this study, the mutant strains of T. onnurineus NA1 strains were developed with excellent formate utilization to enhance H2 production on formate and the function of proteins to be involved in formate metabolism was analyzed. In addition, cultural development was applied to optimize the formate utilization, and as a result, H2 production was increased from increased biomass. Chapter 1 reviews research on formate as a hydrogen energy carrier, formate hydrogen lyase (FHL) systems, formate transporting in formate/nitrate transporter (FNT) families and adaptive evolution. Chapter 2 provides a research on adaptive evolution to improve the strain for enhanced production of H2. T. onnurineus NA1 was grown to stationary phase to trigger spontaneous mutation and then transferred to a fresh medium containing formate as an energy source at 80°C. Through serial transfers, physiological changes were monitored by performing batch cultures and gradual increases in cell density, formate consumption and H2 production were observed. After 156 transfers, the evolved strain (WTF-156T) showed higher values in cell density, formate consumption and H2 production in comparison with the parent strain. In order to obtain an integrative understanding of the genetic and phenotypic changes during evolution on formate, genome sequencing was performed using the PacBio Single Molecule Real-Time (SMRT) sequencing technology. As a result, 11 mutation genes were discovered. When each reverse mutation was introduced in the evolved strain, two revertants for TON_1561 and TON_1573 encoding frhAGB-encoding hydrognenase β subunit and formate transporter, respectively exhibited significant decreases in the cell density and H2 production. Among the mutations identified during adaptive evolution on formate, an effect of a mutation in formate transporter where the 52nd alanine (A) residue was replaced with threonine (T) was investigated. The mutation, when introduced into the genome of the parent strain, conferred increase in formate consumption and H¬2 production. This result confirmed that the TON_1573 (A52T) mutant displayed higher specific formate uptake rate than the wild-type strain in the cell suspension experiment. Another adaptive passage carried out by culturing repeatedly in a bioreactor revealed that a mutation of formate transporter took place at the same residue, indicating that substitution of this residue at the formate transporter may cause a substantial increase in the formate uptake and then enhance cell growth. In order to infer the effect of mutation in TON_1573, the protein structural models were constructed and analyzed through homology modeling using bacterial formate/nitrate transporter (FNT) protein as template. In Chapter 3, the cultural optimization of genetically engineered strains were carried out to increase cell mass and H2 production from formate.
