Human endometrium repeats the endometrial cycle, periodic proliferation, decidualization, and menstruation, in response to the ovarian steroid hormones. This dynamic cycle controls the short-term receptivity of the endometrium for the successful pregnancy. Decidualization of endometrial cells triggered by endometrial stromal cells (EnSCs) is an essential step for the formation of pregnancy environment. Here, the changes in the morphology of intracellular organelles and mitochondrial energy metabolism were investigated according to the cycle of T-hEnSCs, the EnSC line. c-AMP and MPA were treated for 6 days to induce deionization of T-hEnSCs. Decidualized T-hEnSCs display distinct morphological characteristics when compared with proliferative T-hEnSCs, such as ribosome-rich endoplasmic reticulum, increased formation of vesicles, and shorter mitochondria. Next, energy metabolism between decidualized and proliferative T-hEnSCs were compared by measuring the oxygen consumption ratio (OCR). Decidualized T-hEnSCs were lower in the basal respiration and the ATP linked respiration compared with proliferative T-hEnSCs, indicating that decidualized T-hEnSCs were less dependent on OXPHOS than proliferative T-hEnSCs for energy production. Thus, there were observed the distinct characteristic of T-hEnSCs in ultrastructure and energy metabolism according to the cycle of endometrium in in vitro system and found that energy metabolism in T-hEnSCs were correlated with mitochondrial morphology.
