Ethnophamacological relevance: Raphanus sativus L. seed (RSLS) extracts has been used in clinical material for treatment of digestion, chronic cough, anti-microbial, and anti-fungal effects. Moreover, recently it showed cytotoxic to human colon cancer cell. However, the anti-cancer effects and cell death mechanism on head and neck cancers have not been studied. Aim of the study: To evaluate the anti-cancer activity of the RSLS extracts on oral squamous cell carcinoma (OSCC) cells and their cancer stem cells (CSC)-like characteristics acquired cells. Materials and methods: In this study, I investigated the anti-cancer properties of RSLS extracts in KB cells and CSC-liked KBCD133+ cells. Results: RSLS extract induced cell apoptosis in KB and KBCD133+ cells. Moreover, RSLS inhibits nuclear translocation of β-catenin, cell migration and invasion rates. Treatment of RSLS resulted in increase of Axin and it leds to reductions of β-catenin in KBCD133+ cells. Conclusion: Accordingly, these results indicated that RSLS induced apoptosis of OSCC cells through inhibiting the activity of β-catenin involving increase of Axin. RSLS could be a potential anti-cancer candidate for malignant OSCC which acquired CSC-like characteristics.
