Lack of oxygen, commonly known as hypoxia is a major concern in various treatment modalities of tumors, including chemotherapy, photodynamic therapy and radiotherapy. Due to abnormal growth and poor vasculature, most of the solid tumors commonly suffer hypoxic conditions. These conditions lead to the overexpression and stabilization of hypoxia inducible factor-1alpha (HIF-1α) protein in tumor cells. HIF-1α has been correlated with multiple pathways of gene expressions, which results higher resistance and tumor survival. To overcome the conditions of hypoxia and supply cancerous tissues with more oxygen than the partial pressure at tissue level, oxygen based microbubbles have been investigated by various researchers. Microbubbles are naturally echogenic, shelled molecules containing gas core. Microbubbles may also be used for targeted drug/gas delivery due to their behavior of cavitation under high intensity ultrasound. However, microbubbles face size limitations, and therefore, therapeutic applications focus on nanobubbles with similar nature of biocompatible monolayer shells and gaseous cores. Nanobubbles can be synthesized using various shell types including lipids, proteins and polymers and they can be injected intravenously as a source of oxygen to reverse hypoxia and resultantly downregulate HIF-1α. Phospholipids are biocompatible and biodegradable molecules and they can be used for synthesis of stable nanobubbles in the presence of suitable emulsifying agents. Addition of polyethylene glycol (PEG) to the phospholipids enhances the stability, longevity and biocompatibility of the shells. Varying the ratio of PEG-derivatives determine the size, distribution and zeta potential of the nanobubbles. In this research, oxygen nanobubbles (ONBs) were synthesized using phospholipids as shell composition. The ratio of base lipid and emulsifying agents were determined after experiments to obtain optimal size and number of particles. Characterization of ONBs were carried out using optical, fluorescence, confocal and electron microscopy. Size and number of particles were measured using nanoparticle tracking analysis (NTA) and zeta potentials were measured using dynamic light scattering (DLS). The ONBs were then evaluated for their biocompatibility, oxygen delivery, reversal of hypoxia and downregulation of HIF-1α. A novel hypoxia chamber was designed to create hypoxia conditions. Intracellular hypoxia was evaluated through a fluorescent dye. Then these ONBs were used for antitumor and immunosuppressive drug delivery applications. For antitumor drug delivery applications, doxorubicin was encapsulated in ONBs. Dox/ONBs were characterized using microscopy techniques and their mechanism of action was evaluated by reactive oxygen species (ROS) assays. The effectiveness of Dox/ONB was evaluated both in normal and hypoxic conditions to establish the role of ONBs in effective tumor therapy. Cellular uptake of Dox/ONB was evaluated through confocal microscopy imaging. Furthermore, a hydrophobic immunosuppressive drug namely mycophenolic acid (MPA) was encapsulated in ONBs. MPA/ONB were used to reduce cytokine release of IL-2, and TNF-a. In totality, our results suggest that ONBs are biocompatible and they can be used to reverse hypoxic conditions and downregulate HIF-1α. Additionally, these ONBs can be used to encapsulate drugs for antitumor and immunosuppressive applications.
