EGFR-TKI인 gefitinib에 민감하거나 내성을 갖는 비소세포폐암에서 LCB와 Ecn의 EGFR 및 c-MET의 이중 표적 여부를 확인하기 위해, 인산화효소 활성 분석, pull-down 분석, ATP-competition 분석 및 molecular docking simulation을 수행하여 평가하였다. LCB와 Ecn은 EGFR과 c-MET의 ATP-결합 부위에 직접 결합함으로써 인산화효소 활성과 인산화 발현을 억제하였다. 이어서 LCB와 Ecn은 EGFR 및 c-MET의 하위 단백질인 AKT의 인산화를 억제함을 확인하였다. 또한, LCB와 Ecn은 세포증식 뿐만 아니라 콜로니 형성을 유의하게 억제하였다. Western blotting과 유세포분석 결과, 폐암에서 LCB와 Ecn의 처리는 cyclin B1과 cdc2의 발현을 억제하고 p21, p27의 발현은 증가시킴으로써 세포주기의 G2/M 정지를 유도하였다. 세포주기의 sub-G1 비율과 Annexin V 염색은 LCB와 Ecn이 폐암 세포주에서 세포사멸을 유도한다는 것을 보여주었다. 유세포분석 및 western blotting은 LCB와 Ecn이 ROS의 형성, ER stress의 유도 및 MMP의 탈분극을 일으킴으로써 세포사멸과 관련된 단백질들을 조절한다는 것을 확인하였다. 또한, LCB와 Ecn의 처리에 의해 caspase들이 활성화되었고, pan-caspases 억제제인 Z-VAD-FMK의 처리의 결과는 LCB와 Ecn이 caspase-의존적 세포사멸을 유도한다는 것을 나타냈다. LCB와 Ecn은 EGFR과 c-MET를 동시에 표적함으로써 TKI에 대해 민감하거나 내성을 갖는 폐암 세포의 증식을 억제하고 약물 내성을 극복하였다. 다음으로 구강편평상피암 세포주에서 LCC와 LCH는 경쟁적인 ATP와의 결합을 통해 JAK2와 직접 결합함으로써 인산화효소 활성을 차단하였다. LCC와 LCH에 의해 JAK2뿐만 아니라 JAK2의 하위 분자인 Signal transducer and activator of transcription 3 (STAT3)의 인산화 발현을 강하게 억제하는 것으로 나타났다. STAT3의 하위 단백질들의 발현을 관찰한 결과, LCC와 LCH 처리로 인해 Bcl-2, Mcl-1, survivin, cyclin D1은 감소하였고, p21과 p27은 증가하였다. 또한, LCC 및 LCH 처리는 세포증식 및 콜로니의 형성을 유의하게 억제하였다. To date, many molecular target inhibitors have been developed clinically to improve the survival of cancer patients. Some kinase-targeted drugs are in clinical trials. Because tyrosine kinases (TKs) is involved in a wide range of cellular functions, including cell growth, proliferation, differentiation, and immune function, it has been highlighted as a potential and attractive molecular target for cancer therapy. Janus kinase 2 (JAK2), a non-receptor TK, is highly expressed in several cancer cells as a signaling pathway regulator activated by growth factors and cytokines. Epidermal growth factor receptor (EGFR), and c-MET are overexpressed in cancer and involved in cancer growth and progression. EGFR inhibitors (e.g., gefitinib) have been used to treat lung cancer with effective response initially. However, over time, drug resistance is acquired. Strategies to overcome drug resistance can lead to successful cancer treatment. Therefore, there is a need for the development of novel drugs that target TKs. There is a need for ongoing research to develop safe and effective cancer prevention and treatment. Natural plants that have long been used to treat a variety of diseases with great potential. Phytochemicals contained in natural plants are relatively stable and effective molecules. They are known to have anti-tumor activity in various cancer cells. Licorice (Glycyrrhiza inflata) is one of the oldest and most used botanicals in Chinese medicine. The root of G. inflata has various biological activities such as anti-oxidant, anti-inflammatory, anti-viral, and anti-tumor effects. Licochalcone (LC) and Echinatin (Ecn) isolated from licorice root or rhizome are a type of retrochalcone. Until now, LCA has shown anti-cancer efficacy in various cancers in many studies. However, studies on the effect of other LCs and Ecn are insufficient. Thus, in this study, we examined effects of LCB, LCC, LCH, and Ecn by targeting tyrosine kinase in lung and oral cancer cells. First, we evaluated TKs targets of LCB and Ecn in gefitinib-sensitive or -resistant lung cancer by performing pull-down assay, ATP-competitive assay, kinase assay, and molecular docking simulation. LCB and Ecn inhibited kinase activity and phosphorylation by binding directly to ATP-binding pockets of EGFR and c-MET. LCB and Ecn then inhibited phosphorylation of AKT, downstream proteins of EGFR and c-MET. LCB and Ecn significantly inhibited colony formation as well as cell proliferation in cell lines sensitive or resistant to gefitinib. Western blotting and flow cytometry analysis showed that treatment with LCB and Ecn in lung cancer cells inhibited the expression of cyclin B1 and cdc2 but increased the expression of p21 and p27, leading to G2/M arrest in the cell cycle. Sub-G1 phase and Annexin V staining showed that LCB and Ecn induced apoptosis in lung cancer cell lines. Further flow cytometry and western blotting confirmed that LCB and Ecn induced ROS formation and ER stress and depolarized MMP to regulate apoptosis-related proteins. In addition, caspases were activated by treatment with LCB and Ecn. Cell proliferation inhibited by LCB and Ecn was partially recovered by treatment with Z-VAD-FMK, a caspase inhibitor. Next, LCC and LCH blocked kinase activity by directly binding to JAK2 in competition with ATP in oral cancer cell lines. LCC and LCH strongly inhibited the expression of phosphorylation of signal transducer and activator of transcription 3 (STAT3), a downstream molecule of JAK2, as well as JAK2. Results on the expression of downstream proteins of STAT3 showed that levels of Bcl-2, Mcl-1, survivin, and cyclin D1 were decreased while those of p21 and p27 were increased due to LCC and LCH treatment. In addition, LCC and LCH treatment significantly inhibited cell proliferation and colony formation. Sub-G1 and Annexin V staining showed an increase in the proportion of apoptotic cells. DAPI staining indicated the induction of apoptosis by showing nuclear fragmentation and chromatin condensation. To evaluate the apoptosis mechanism of LCC and LCH in oral cancer cells, flow cytometry and western blotting were performed. Results showed that ROS accumulation, MMP loss, caspases activation, and apoptosis-related proteins were regulated. In conclusion, LCB, LCC, LCH, and Ecn as naturally derived components can induce apoptosis in lung and oral cancer cells by targeting TKs (EGFR, c-MET or JAK2). Therefore, LCB, LCC, LCH, and Ecn have potential as TKIs. They could be used as promising new therapeutic agents or adjuvants for human cancer treatment.
