Accordingly, sepsis is a syndrome of life-threating multiple organ dysfunction caused by an improper host reaction to infection. Sepsis is categorized into sepsis, severe sepsis, and septic shock according to severity of reaction. Several microorganisms were reported to cause sepsis, including gram-negative bacteria, gram-positive bacteria, some fungi species, some viral infections, and some parasites. Among them, Escherichia coli and Staphylococcus aureus are the leading causes of sepsis. Several epidemiological studies were performed to report the incidence and mortality in human and animals. In human, mortality estimated to be between 17~26% of sepsis and severe sepsis, respectively. Numerous experimental models were utilized to study mechanisms, pathogenicity and treatment of sepsis. The models include bacterial injections, cecal slurry injection, lipopolysaccharide (LPS) injection, and others. LPS, the main part of outer membrane of gram-negative bacteria is widely injected to rodents to induce sepsis. Sepsis causes severe disturbances range from mild fever, induction of cytokines, pro-inflammatory reaction, hypomagnesemia, high lactate level and organs dysfunction. The treatment of sepsis remains a challenge. Therefore, there are alternative treatments, such as medicinal plants and non-chemical (physical) therapies. Diabetes is defined as inability of pancreases to produce enough insulin to reduce the level of glucose due to genetic or environmental factors affecting beta cells. Hyperlipidemia is a marked increase in the levels of lipids, mainly low-density lipoprotein cholesterol, triglycerides, and notable decrease in high-density lipoprotein levels. Diabetes is considered as a leading cause of death in the world, it affects about 10% of global population while hyperlipidemia cause more than 50% of cardiac ischemia worldwide. Treatment of diabetes and hyperlipidemia remain a challenge because the treatment protocol must be for life. These long journeys of treatment cause a burden on patients and their families, besides the complications, which may experience. The objectives of this study are 1) to investigate the pharmacologic effects of alcohol extract (AE) and water extract (WE) of Korean red ginseng (KRG) on LPS-induced sepsis in rat model and raw 264.7 macrophage cells concerning organs injuries, blood pressure, acid-base balance, blood metabolites, cytokines, oxidative stress, MAPK pathway signaling, and p65 translocation, 2) to investigate the effects of KGR on the alloxan-induced diabetes and poloxamer 407-induced hyperlipidemia after long-term oral intake, 3) to examine the pharmacological activities of ultraviolent blood irradiation (UBI) on LPS-induced sepsis in rabbit model with respect to blood metabolites, liver, lung, and kidney function, lactate level, magnesium condition, acid-base balance, cytokines, pro-inflammatory reaction, and oxidative stress, and 4) to investigate the effects pharmacological and biochemical of UBI on diabetes and hyperlipidemia.
Part 1. Korean red ginseng extract reduces inflammatory response via suppression MAPK signaling and p65 nuclear proteins translocation in rats and RAW 264.7 macrophage cells exposed to Lipopolysaccharide Lipopolysaccharides (LPS) cause systemic inflammatory responses, which are characterized by high mortality and multiple signs, including metabolic disturbances, respiratory acidosis, hypotension, and vital organs disorder. Cytokines secretion and oxidative stress are the main features of the disease. Diagnosis and treatment of systemic inflammation remain as a challenge to researchers. KRG is one of medicinal herbs that showed a potent antioxidant effect. We aimed to study the protective effects of KRG on systemic inflammatory response in rats and RAW 264.7 macrophage cells induced by LPS. The rats were treated with water and alcohol extracts of KRG for four weeks to prevent the inflammatory response. The result showed that LPS toxin increased morbidity and mortality, and induced liver, kidney, and lung injuries manifested by deteriorated biomarkers. Hypotension, hypomagnesemia, acidosis, and oxidative stress were observed in septic rats. However, KRG extracts attenuated liver, kidney, and lung enzymes and metabolites in treated groups via its anti-inflammatory and antioxidant properties. Furthermore, KRG improved magnesium and blood pressure in the treated groups. RAW 264.7 macrophage cells exposed to LPS disturbance in translocation of p65 and MAPK/p38. Nevertheless, KRG-pretreated cells did not significantly alter. In conclusion, KRG reduced the rates of mortality and morbidity of treated rats — liver, kidney, and lung injuries were protected in the treated groups through the potentiation of antioxidant defense. KRG was able to conserve mitochondrial function, inhibiting the activation of MAPK/p38 signaling and suppressing NF-κB p65 cytoplasm-nucleus transport. Further studies are needed to examine the effects on chronic conditions in animal models and human.
Part 2. Long-term oral intake of Korean red ginseng improves hypomagnesemia, hyperlactatemia, base deficit, and metabolic acidosis in an alloxan-induced diabetes and poloxamer 407-induced hyperlipidaemia rabbit models KRG is widely used for its various pharmacological activities, including effects on diabetes and its complications, such as hyperlipidemia (HL). This study aims to investigate the effect of KRG on mortality-related hypomagnesemia, hyperlactatemia, metabolic acidosis, and other diabetes- and HL-induced abnormalities. Type 1 diabetes was induced by IV injection of alloxan monohydrate 110 mg/kg into New Zealand white rabbits weighing 2~2.5 kg; HL was induced by injecting poloxamer 407 subcutaneously. KRG was supplied in drinking water for 20 weeks. The effects of the KRG treatment on diabetes were evaluated through hematological and biochemical analysis, including ELISA assays for insulin and glycated hemoglobin A1c (HBA1c) before and after KRG extract was supplied. The serum glucose, insulin, and HBA1c levels were significantly improved after the KRG treatment compared to those found before KRG treatment. In addition, Mg2+, lactate, and base deficit, and acidosis was significantly enhanced in treated rabbits. Moreover, KRG showed hypolipidemic and hepato- and reno-protective effects. Likewise, Electrolytes and blood gases conditions were improved. The biochemical and hematological analysis data demonstrate that the KRG is effective to alleviate diabetes and HL serious signs.

Ultraviolet-c haematogenous oxidation therapy of lipopolysaccharide-induced endotoxemia in a rabbit model: A biochemical study Systemic inflammatory reaction due to severe response to toxins of infection associated with immune inhibition leading to multi-organ dysfunctions and high mortality. Prototype treatment can support but, is not considered a radical treatment. Ultraviolet (UV) blood irradiation produces bio-photons in the body, have therapeutic effects when moving across the cells. The aim of this study is to evaluate the effects of UV-c Haematogenous Oxidation Therapy (HOT) in lipopolysaccharide (LPS)-induced endotoxemia rabbit model. A total of 40 rabbits randomly divided into four groups including normal control (NC). LPS and LPS+UV-c HOT groups received 0.1 mg/Kg LPS toxin of E. coli, UV-c HOT and LPS+UV-c HOT groups subjected to UV-c HOT treatments weekly for 5 times. Blood collected, perfused with oxygen, UV-c directly irradiated into blood and auto-transfused. Rabbits were sacrificed after 5 weeks; blood and serum were collected for analysis. Survival rate, liver, kidney, lipid profile, and blood ions were assessed in UV-c HOT treated rabbits. Four rabbits were died in the LPS group whereas, other groups recorded no death. UV-c HOT modulated blood parameters, cytokines, liver enzymes, lipoprotein metabolism and electrolytes homeostasis. Collectively, these results propose that UV-c HOT performance for human and animal endotoxemic or septic patients can be evaluated and considered.
Part 4. Effect of haematogenous oxidation therapy with ultraviolet-c irradiation on alloxan-induced diabetes and poloxamer 407-induced hyperlipidemia in rabbits

The study evaluates the effects of HOT on the blood, when a low dose of Ultraviolet-C (UV-c) is directly irradiated to the blood in a diabetic rabbit model and to evaluate the effects of treatment on diabetic and hyperlipidemic rabbits. Type 1 Diabetes and hyperlipidemia (HL) were induced by intravenous injection of alloxan monohydrate and subcutaneous injection of poloxamer 407, respectively. A 10 ml blood was collected from diabetic rabbits, blood was being perfused with oxygen for 10 seconds and UV-c was irradiated to the blood, UV-irradiated blood was transfused back to the original rabbits. The HOT treatment was performed a total of 10 times. It was evaluated the effects of the HOT treatment on diabetes and HL through hematological and biochemical analyses before and after HOT treatment were performed. The results indicated that the reduced body weight was increased, and blood glucose levels were significantly reduced after the HOT treatment was performed when compared to those before the HOT treatment. Besides, creatinine, blood urea nitrogen and uric acid levels indicating renal functions were significantly reduced when compared to those prior to the HOT treatment. When the HOT treatment was performed in a diabetic and HL rabbit model, our results indicate that blood glucose levels and lipids profile were significantly improved.
