The purpose of this experiment is to investigate the functional role of panobinostat in gastric cancer cells. Gastric cancer is the fifth most common cancer and the third leading cause of cancer-related deaths worldwide. The histone deacetylase inhibitors are a new class of cytostatic agents for various cancers and diseases. Panobinostat (LBH-589, Novartis) is a pan-HDAC inhibitor approved by the FDA for multiple myeloma. Despite many clinical and pre-clinical trials that have been conducted to assess the anti-cancer effects of panobinostat, only a few reports were investigated the role of panobinostat on gastric cancer. In this study, panobinostat significantly inhibited cell viability and proliferation of SNU484 and SNU638 cells in a dose-dependent manner. After panobinostat treatment cell viability and colony-forming ability significantly diminished. Panobinostat treatment induced apoptosis as indicated by increased protein levels of cleaved-poly ADP-ribose polymerase (PARP), cleaved-caspase 3, increased sub G1 phase and increased Annexin V positive cell rate in SNU484 and SNU638 cells. Panobinostat treatment arrested G2/M phase of SNU484 and SNU638 cells, which indicated by FACS analysis and decreased the G2/M phase regulatory-associated protein levels of P-Wee1, Myt1, and Cdc2. Also, panobinostat treatment significantly inhibited the metastasis of SNU484 and SNU638 cells by targeting MMP 9 and E-cadherin. Besides, panobinostat treatment decreased the protein levels of p-Akt and FOXM1. These results indicate that panobinostat inhibits proliferation, metastasis, and cell cycle progression of gastric cancer cells through the apoptosis induction and inactivation of Akt/FOXM1 signaling. Taken together, our present study suggests panobinostat is a potential anti-cancer reagent for the treatment of gastric cancer.
