1. Highly Sensitive Impedimetric DNA Sensor on a Diazonium-Modified Gold Electrode Prepared by Electrochemically Selective Reduction
The development of a simple, rapid, and reliable method for detecting the virus at very low levels is an important objective in useful biosensors. To that end, a gold surface was electrochemically modified by reducing 4-nitrobenzenediazonium tetrafluoroborate. Radical formation and its polymerization were significantly affected by the reaction temperature, the concentration of the diazonium precursor solution, the number of cyclic voltammetry (CV) cycles, and the CV scan rate. Two cycles at 50 mV/s and three cycles at 100 mV/s in 0.1 mM 4-nitrobenzenediazonium at 5 °C were optimized to be experimental conditions for monolayer surface coverage. Because monolayer allows the constant distance between the electrode surface and the target DNA, it is the most capable surface coverage for the high performance of an impedimetric DNA sensor. And then, the nitro functional group was electrochemically converted into the amino functional group. Glutaraldehyde was used to link probe DNA to the amino functional group to detect the target DNA of the dengue virus, after which the immobilized probe DNA was hybridized with various concentrations of target DNA. Electrochemical characterizations during each step in the overall transformation, including diazonium functionalization, converting from nitro groups to amino groups, glutaraldehyde treatment, probe DNA immobilization, and target DNA hybridization, were monitored by electrochemical impedance spectroscopy and CV in ferri/ferrocyanide solution.
