In the present study, CD38, an important regulator of NAD-dependent deacetylase, such as SIRT1, which regulation senescence and energy metabolism in mammalian cells, was investigated. The main enzyme activity of CD38 is NADase, and NAD is used as a substrate of CD38 for the synthesis of cADPR and is involved in the regulation of NAD levels in cells. However, direct evidence linking CD38 itself with the observed senescence effect in vitro and relevant molecular mechanisms are still deficient. In this study, investigated whether the expression of CD38 induced cellular senescence of WI38 cells. In addition, examined the anti-senescence effect of latifolin and fisetin in H2O2-induced cellular senescence. The production of cADPR increased CD38-dependently, and it was confirmed that the NAD levels and SIRT1 activity in cells were significantly decreased. It was confirmed that the expression of SA-β-gal positive cells and senescence-related protein was increased by CD38, and was decreased when CD38 was inhibited by siRNA. Confirmed that the cellular senescence by H2O2 was decreased by CD38 inhibition, and the expression and activity of SIRT1 was increased and the expression of Ac-p53 was decreased. In addition, confirmed that CD38 induced cellular senescence in MEFs. Taken together, the demonstrate that CD38 effectively inhibits the activity of SIRT1 by reducing NAD+ levels and effectively induces cellular senescence by increased senescence-related biomarkers.
