본 연구는 다년생 식물로서 이탈리아 요리에 많이 사용되는 식용식물인 Eruca sativa를 70% ethanol로 추출(ES)한 후 n-hexane, ethyl acetate, chloroform, n-butyl alcohol 및 water 용매 극성별로 분획(EHex, EEA, ECHCl3, EBuOH 및 EDW)하여 항염효과를 규명하고 화장품 소재로의 응용성을 연구하였다. ES, EHex, EEA, ECHCl3, EBuOH 및 EDW의 항산화 효능을 평가하기 위해 DPPH 라디칼 소거능을 측정한 결과 EEA와 EBuOH에서 가장 높은 항산화 효과를 보였으며, 총 폴리페놀과 총 플라보노이드의 함량도 EEA와 EBuOH에서 가장 높게 나타났다. Mushroom tyrosinase 활성은 EEA가 가장 유의하게 저해하였으며, B16F10 melanoma 세포 내에서의 ES, EEA 및 EBuOH를 처리하여 멜라닌 생성 저해를 확인한 결과 ES, EEA 및 EBuOH 모두에서 멜라닌 생성을 저해하였으며 특히 EEA가 가장 효과가 뛰어났다. Lipopolysaccharide (LPS)로 유도된 RAW 264.7 대식세포에서 염증매개물질인 nitric oxide (NO)와 전염증성 사이토카인(pro-inflammatory cytokine) IL-6, TNF-α 생성 저해를 확인한 결과 EEA가 가장 효과가 뛰어났으며, NO를 생성하는 inducible nitric oxide synthase (iNOS)의 mRNA 및 단백질 발현을 유의하게 감소시켰다. 또한, LPS는 reactive oxygen species (ROS)를 생성하여 염증을 유발시키는데 EEA는 LPS를 처리한 RAW 264.7 세포에서 생성된 ROS를 제거하는 항산화 효과를 나타냈다. HPLC 분석한 결과 EEA에서 isoquercitrin이 다량 검출되었으며 quercetin, kaempferol 및 isorhamnetin이 소량 검출되었다. Isoquercitrin은 NO 생성 저해에서 IC50이 1.81 μM으로 효과가 뛰어났다. E. sativa 추출물(ES)을 토너, 에멀션 제형에 0.05, 0.1%로 적용하여 일광, -20, 4, 25, 55 ℃에서 4 개월간 안정성을 평가한 결과 pH 및 점도에 대한 영향은 크게 없었으나 대조군 및 ES를 첨가한 에멀션 55 ℃에서 분리가 일어나는 것을 확인하였다. 아토피 피부염과 여드름을 일으키는 Staphylococcus aureus, Propionibacterium acnes 외 6가지 세균, 진균에 대해 ES, EHex, EEA, ECHCl3, EBuOH 및 EDW의 항균력과 방부력 시험을 평가한 결과 EEA는 8종의 균 모두 효과적으로 항균력을 나타냈으며, ECHCl3는 Pseudomonas aeruginosa를 제외한 모든 균에 대해 항균력을 보였다. ES를 0.05, 0.1%를 함유하여 제조한 토너와 에멀션, 대조군 제형의 방부력 시험에서 합격하여 방부력을 증가시키는 효과를 보일 것으로 확인된다. Franz diffusion cell을 이용하여 피부침투시험을 평가한 결과 EEA 처리 시 1, 3, 6 h에서 0.022, 0.048, 0.058%가 침투되어 실제 사용 시 임상적인 효능이 있을 것으로 예측할 수 있다. 위 결과를 통해 EA 분획물은 iNOS의 mRNA 및 단백질 발현량을 감소시켜 NO 생성과 IL-6, TNF-α 생성 저해함으로써 항염증 효과를 가지며, 이는 HPLC 분석과 항염 결과를 통해 플라보노이드인 isoquercitrin의 효과로 확인된다. 추가적으로 염증반응 후 과색소침착증(post-inflammatory hyperpig- mentation, PIH) 개선 및 화장품 제형의 변질을 막을 수 있는 천연보존제 역할을 할 수 있을 것으로 판단된다. 즉, 천연소재 E. sativa 추출물은 염증 완화를 통해 최근 증가하고 있는 아토피 피부염, 민감성 피부를 포함한 만성 염증성 피부질환 개선과 자극 완화 화장품 소재로서 활용가치가 우수한 것으로 사료된다.

Then ES was fractionated using n-hexane, chloroform, ethyl acetate, n-butyl alcohol and water (EHex, EEA, ECHCl3, EBuOH and EDW). DPPH radical scavenging activity, total polyphenol and total flavonoid were evaluated by 70% ethanol extract and solvent fractions (ES, EHex, EEA, ECHCl3, EBuOH and EDW). As a result, EEA and EBuOH showed the highest contents and anti-oxidant effect. EEA showed mushroom tyrosjnase inhibitory activity. ES, EEA and EBuOH showed an inhibitory effect of melanin production in B16F10 melanoma cell. As a result, ES is expected to have skin whitening efficacy. The anti-inflammation effects of ES, EEA and EBuOH on the production of nitric oxide (NO), inducible NO (iNOS) and pro-inflammatory cytokines IL-6 and TNF-α in lipopolysaccharide (LPS)-induced RAW 264.7 cells were measured. EEA inhibited NO production in a dose dependent manner and significantly decreased the mRNA and protein expression of iNOS. In addition, EEA effectively inhibited reactive oxygen species (ROS) causing inflammation at the cellular level. It was confirmed by HPLC analysis that the EEA contained a large amount of isoquercitrin and quercetin, kaempferol and isorhamnetin were detected in small amounts. Isoquercitrin was found to have a very low IC50 (1.8 μM) in inhibition of NO production. E. sativa extract (ES) was applied to 0.05, 0.1% the toner and emulsion cosmetics formulation to test the stability. Skin penetration rate of ES was evaluated using Franz diffusion cell and then, ES can be predicted to have a clinical efficacy.
