UV 노출은 활성산소종 (Reactive oxygen species, ROS)의 생성을 유도하여 피부 광노화를 유발한다. 피부 광노화는 피부장벽기능감소, 거칠기, 건조함을 특징으로 한다. 본 연구에서는 흑미 및 블루베리 혼합 발효산물이 UVB로 조사된 인간각질형성세포 (HaCaT keratinocytes)에서 광노화와 관련된 기전인 apoptosis에 미치는 영향을 조사하여 작용기전을 결정하고, 동물모델에서 피부수화와 피부장벽기능과 같은 노화관련 파라미터를 측정함으로써 UVB에 의해 유발된 광노화에 대한 발효산물의 보호효과를 평가하였다. Cell-free system에서 발효산물의 항산화효능을 평가한 결과, free radical 소거 활성과 환원력이 농도의존적으로 증가되었음을 알 수 있었으며, 산화적 DNA 손상에 대해 보호효과가 있음을 나타냈다. HaCaT 세포에서 UVB의 조사는 ROS 생성과 apoptosis를 유도하였으며, 발효산물의 처리에 의해 ROS 생성 수준은 감소되었고 caspasecascade 경로를 막음으로써 apoptosis 유발을 억제하였음을 확인하였다. 또한, 흑미 및 블루베리 혼합 발효산물은 HaCaT 세포 및 hairless mice에서 FLG, TGM 및 COL1A1과 같은 피부 장벽 기능에 관여하는 유전자의 발현을 증가시켰으며, UVB로 광노화를 유발한 hairless mice에서 등 피부의 표피 두께와 수화를 개선시켰다. 이러한 결과는 흑미 및 블루베리 혼합 발효 산물이 UVB로 유도된 광노화에 대해 피부장벽기능과 보습을 개선함으로써 피부보호효과가 있음을 나타낸다. 따라서 흑미 및 블루베리 혼합 발효산물은 UVB로 인한 피부 광노화를 개선하는데 유익한 효능을 갖는 건강기능식품 소재가 될 수 있을 것으로 사료된다.

UV exposure induces reactive oxygen species (ROS) production, causing skin photoaging. Skin photoaging is characterized by decreased skin barrier function, roughness and dryness. In this study, the mechanism of photoaging was determined by investigating the effect of BBL on apoptosis associated with photoaging in UVB-irradiated HaCaT keratinocytes, and the protective effect of BBL against UVB-induced skin photoaging was evaluated by measuring aging-related parameters such as skin hydration and skin barrier function in animal model. As a result of evaluating the antioxidant activity in cell-free system, BBL showed a dose dependent increase in free radical scavenging activity, reducing power, and protective activity against oxidative DNA damage. Intracellular antioxidant activity and anti-apoptotic effect were evaluated in UVB-irradiated HaCaT cells. BBL inhibited intracellular ROS generation and apoptosis induced by UVB irradiation, and it was confirmed that apoptosis was suppressed through inhibition of the caspase cascade pathway. In addition, BBL increased the expression of genes involved in skin barrier function such as FLG, TGM, and COL1A1 in HaCaT cells and hairless mice. Oral administration of BBL improved the epidermal thickness and hydration of the dorsal skin in UVB-irradiated hairless mice. These results indicate that BBL has a protective effect by improving skin barrier function and moisturization against UVB-induced photoaging. Thus, it suggests that BBL can be used as a health functional food with beneficial effects on UVB-induced skin photoaging.
