In this study, the eutectic temperature and ratio of deep eutectic solvents (DES) based on choline chloride and dicarboxylic acid were determined and synthesized. Depending on the number of carbon atoms of hydrogen bond donor (HBD), the difference between the eutectic temperature and the ratio was confirmed, and when the number of carbon atoms is odd, it has a lower eutectic temperature and exists as a transparent liquid at room temperature. In addition, rutin, hyperoside, and isoquercitrin, which are flavonoid components of Pyrus ussuriensis leaves, were identified through high performance liquid chromatography (HPLC) and HPLC-MS/MS analysis. DES-4, which showed higher extraction efficiency than conventional solvents, was selected and the solvent characteristics of DES were evaluated through response surface methodology (RSM) based application of extraction of Pyrus ussuriensis leaves. The predicted optimum extraction conditions are 30°C, 30.85 wt% DES, stirring speed of 1113 rpm, extraction for 1 hour, and a flavonoid content of 217.56 μg/ml can be expected, the result of three reproducibility experiments, high significance with a confidence range of 0.05 was confirmed at 223.37 μg/ml, 219.10 μg/ml and 227.11 μg/ml. The antioxidant activity of the extract extracted with DES was evaluated through DPPH assay and ABTS assay, and 209.58 ppm of extract showed an antioxidant activity equivalent to 75.63 ppm of L-ascorbic acid, and it was confirmed that the activity did not decrease and preserved when extracting DES.
