However, a syngas fermentation process that produces bioethanol using CO-based syngas could be used to overcome this problem. In this study, I developed the technology for an economical syngas fermentation process by optimizing culture conditions to improve ethanol productivity, reducing the cost of the medium, and increasing gas-liquid mass transfer using a sintered sparger. Optimizing the medium by replacing the MES buffer with a potassium phosphate buffer, which occupied the highest medium, reduced the cost by approximately 73%. In addition, optimizing the concentration of NH4Cl increased ethanol productivity by up to 400%. Cell activity was maintained via the addition of MSG. The sintered metal sparger replaced a ring sparger, which increased the kLa to 660% and thus improved mass transfer. A 5 L scale reactor was operated for 40 days based on the optimized conditions and the related process parameters were obtained. The ethanol titer and productivity were 17 g/L and 0.56 g/L/h, respectively. Following this, a 20 L reactor was run based on the process parameter data obtained from the 5 L scale reactor. To operate the 20 L fermenter, I created a clean in place (CIP) and operation protocols. In addition, I modified the sterilization system to prevent microbial strains from becoming contaminated during cultivation. Furthermore, cultivation was possible in actual syngas, i.e., coal-based gas derived from the IGCC of Korea Western Power Plant as a feed gas. Operating the 20 L bioreactor under the optimized experimental conditions resulted in an ethanol titer and productivity rate of 23 g/L and 0.43 g/L/h, respectively.
