Microsatellite marker를 이용한 집단의 유전적 구조 분석 결과 제주와 남해안아구계 집단 그리고 남부 및 중부아구계 집단이 각각 서로 독립된 분기군을 형성하는 것을 보여주었다. 한편 엽록체 DNA marker를 통한 단상형의 변이를 토대로 network를 제작하여 지리적 분포를 확인한 결과 지역과는 무관하게 단상형이 퍼져있는 것을 확인하였다. 결과적으로 microsatellite marker 분석을 통해 제주와 남해안아구계에 자생하는 새끼노루귀, 남부 및 중부아구계에 자생하는 노루귀는 뚜렷이 구별되지만, 엽록체 DNA marker 분석을 통해서는 구별되지 않았다. 이는 새끼노루귀와 노루귀가 계통적으로 완전히 분화되지 않았다는 것으로 해석될 수 있다. 엽록체에 비해 핵 DNA상의 변이가 상대적으로 더 빠르다는 점에 비추어 두 분류군의 유전적 분기를 추정컨대, 상대적으로 최근 만빙기에 각각이 독립된 피난처를 형성했던 시기부터 시작되었을 것이라 여겨진다. 따라서 새끼노루귀는 노루귀와 별개의 종이 아닌 노루귀의 종하분류군으로 취급되는 것이 타당할 것으로 보인다.

The genus, Hepatica (Ranunculaceae) is widely distributed in Asia, North America, and Europe. There are three species of plants in the Korean Peninsula: Hepatica asiatica Nakai, Hepatica insularis Nakai, and Hepatica maxima Nakai. Among them, H. insularis is smaller than Hepatica asiatica and is mainly distributed in Jeju Island and the southern coastal regions of the Korean Peninsula. However, H. insularis does not differ significantly in morphological characteristics from H. asiatica. Thus, taxonomic entity of H. insularis has been controversial. In this study, we discuss their taxonomic entity by identifying the genetic characteristics of H. insularis using nuclear microsatellite markers and chloroplast markers. We first developed a total of eight microsatellite markers that can be applied to two taxa, considered as H. insularis and H. asiatica. Chloroplast DNA markers were developed for three regions: rps4-rps16, atpE-rbCL, and ndhG-ndhI. These markers were applied to 294 leaf samples of H. insularis (184 individuals in JeJu, Geoje, Sinan, Jangseong, Taean) and H. asiatica (110 individuals in Chungnam, Daegu, Yeongwol, Seorak mountain, Bukhan mountain) from 13 populations. As a result, the microsatellite markers analysis revealed that H. insularis and H. asiatica is genetically distinct. However, the chloroplast DNA markers analysis did not distinguish the two. This can be interpreted as an incomplete lineage sorting. Given that nuclear DNA mutations are relatively faster than chloroplasts, speciation of the two taxa are presumed to have begun at a relatively recent time when each formed an separate refugia during the LGM (Last Glacial Maximum). Therefore, it seems reasonable that H. insularis is treated as a subclass of H. asiatica, not a separate species from H. asiatica.
