In particular, epithelial-mesenchymal transition (EMT) has been demonstrated a critical process whereby epithelial cells undergo molecular changes to acquire mesenchymal phenotypes through upregulation of EMT-inducing transcription factors. It was previously showed that ZEB2, through cooperation with the transcription factor Sp1, plays an important role in the transcriptional upregulation of mesenchymal genes such as integrin α5 and vimentin during cancer cell invasion. Here, I show that ZEB2 cooperates with Sp1 for promoting not only EMT and invasion but also diverse cellular functions during metastasis. ZEB2 induces expression of Sp1-regulated genes such as VEGF, survivin, bcl-2 and cyclin D1 in a Sp1-dependent manner. Collaboration between ZEB2 and Sp1 induces cancer cell survival and proliferation, endothelial cell activation, and tumor angiogenesis. Furthermore, Sp1 stabilizes ZEB2 protein, supporting the presence of a positive feedback loop between ZEB2 and Sp1. Clinical data show that ZEB2 expression was correlated to Sp1 expression, and that those factors were a crucial for predicting survival in cancer patients. Meanwhile, SUMOylation regulates transcriptional activity of many of the transcription factors in transition between epithelial and mesenchymal state. Recently, it was reported that ZEB2 could be SUMOylated at Lys391 and Lys866 and the SUMOylation of ZEB2 reduced its function as a transcriptional repressor of E-cadherin. In this study, I show that lack of ZEB2 SUMOylation reduced expression of genes which associated with EMT and survival and decreased ability of cell invasion and anoikis resistance. To explore whether EMT markers are involved in immune evasion of cancer cells, the effect of ZEB2, Twist1 and Snail on regulation of PD-L1, CCL2 and CD47 was verified in cancer cells and clinical data of patient. Interestingly, knock-down of ZEB2 reduced activity of T cell in Jurkat cells and migration of Thp-1 monocyte cell line. These findings demonstrate cooperative roles of transcription factors and function of ZEB2 SUMOylation in cancer metastasis and immune evasion. The transmembrane four L6 family member 5 (TM4SF5) is highly expressed in a various cancers, including liver, colon, pancreatic cancers. Ab27, a TM4SF5 targeting monoclonal antibody, was validated for liver and colorectal cancer therapy. TM4SF5-specific binding of Ab27 was confirmed in FACS, IF, WB and BIACORE analysis. Ab27 efficiently inhibited tumor growth and targeted tumor in cancer xenograft models. In addition, combination treatment of Ab27 with Sorafenib or Cetuximab enhanced suppression of p-Fak, p-Erk and p-STAT3 expression in cancer cells and inhibited tumor growth in cancer xenograft models. These results verified the efficacy of TM4SF5-targeting antibody and presented novel anti-cancer therapy for liver and colon cancer patients.
