In this study, we conducted an anti-aging efficacy test of coenzyme Q10 by culturing a human-derived skin model composed of the epidermis and dermis using Pumpless Skin-on-a-chip. Coenzyme Q10 is added to tissue equivalents using culture medium, and the platform fabricated uses gravity-based skin-on-a that allows the culture to circulate at the desired flow rate without a pump and external tube. The number of fibroblasts in the dermis layer was compared according to the concentration of the added drug, the differentiation of the skin structure was analyzed, and the tissue contraction rate depending on the added drug concentration was confirmed. The efficacy of coenzyme Q10 was investigated by real-time qPCR and immunohistochemical staining. The expression level of TGF-beta 1, which is one of the paracrine signals that interact with collagen, was confirmed through ELISA analysis. Coenzyme is expected to be effective in suppressing the aging response, including increasing the expression level of proteins required to maintain the barrier function of the skin, and this result can improve preclinical evaluation. It is expected to help promote the use of the Pumpless Skin -On-a-chip system to evaluate the efficacy of coenzyme Q10 added to cosmetics. In this study, it was confirmed whether the skin equivalent showed the same aging state as the actual human body with or without regular compression stimulation during the 28-day culture. Skin aging was implemented using a method of applying a 12-hour stimulus via a compression stimulus drive and resting for 12 hours. H & E staining was used to confirm changes in the thickness of the epidermal layer with and without compression stimulation, and real-time qPCR was used to confirm an increase in the gene expression level of P16, which is an aging marker, by compression stimulation. In conclusion, a decrease in the thickness of the epidermal layer and an increase in the P16 gene observed in skin aging during compression stimulation were observed, and a skin equivalent cultured under the condition of applying compression stimulation was suitable as a skin aging model. It seems that it is useful for testing drugs and cosmetics to reproduce HSE with characteristics equivalent to actual skin aging in the production of these human-derived skin aging models within a period of 28 days. Alternatives to animal experiments for welfare are also expected to be suitable.
