표면 개질을 통해 생체재료의 물리적, 화학적 특성을 제어하고, 생물학적 기능이나 약리적 기능을 강화시키는 연구는 다양한 분야에서 끊임없이 수행되고 발전해오고 있다. 표면 개질의 다양한 방법 중에서, 고분자의 물리적인 흡착으로 형성되는 다층 나노필름을 이용한 방법은 필름 구성 물질이나 기판에 제한이 적고, 구조 및 기능의 정교한 조절이 가능하다는 장점이 있다. 본 연구에서는 고분자 나노필름의 성분 및 구조를 조절하여 생체재료의 기능을 개선하는 전략을 제안한다. 먼저, 사용된 물질과 실험 조건에 따라 변하는 나노필름의 특성을 최근 주목받고 있는 데이터 과학 관점에서 분석하고자 하였다. 구체적으로, 나노필름 제작 실험에서 도출되는 각종 수치들을 데이터로 변환하고, 이를 구성하는 변수들의 관계를 회귀 분석을 통해 추정하는 방식으로 실험 결과를 예측할 수 있는 모델을 제작하였다. 해당 전략은 추후 고분자 나노필름을 기반으로 한 각종 실험의 효율을 향상시키는데 기여할 것으로 예상된다. 또한, 거대 생체 분자인 성장인자의 방출을 제어하기 위한 나노필름을 개발하였다. 삼중층 구조의 나노필름을 통해 성장인자의 전달 효율을 증가시킬 수 있었고, 분해 속도가 상이한 두 고분자들의 블렌딩 비율을 조절하여 방출 속도를 제어할 수 있었다. 해당 나노필름을 통해 3일간 배양액 내 섬유아세포성장인자의 농도를 유지함으로써 유도만능줄기세포 배양 효율이 향상됨을 확인하였다. 마지막으로, 나노필름을 세포막에 코팅하여 세포의 분화를 제어하거나, 기능을 향상시킬 수 있는 기능성 세포 환경을 구현하였다. 연골 분화를 촉진하는 고분자인 피브로넥틴 및 콘드로이틴이황산으로 구성된 나노필름으로 중간엽줄기세포의 연골 분화 효율을 향상시켰으며, 콜라겐과 알긴산으로 이루어진 높은 경도의 나노필름으로 조골 분화 효율을 향상시켰다. 한편, 세포 사멸을 억제하는 물질을 포함한 나노필름을 유도만능줄기세포 표면에 코팅하여, 유도만능줄기세포 전분화능을 유지시킬 수 있었으며, 세포-세포 인력을 촉진하는 나노필름을 제조하여 유도만능줄기세포로부터 형성된 배아체의 분화능을 강화하였다. 본 연구의 결과들은 세포 배양 환경 또는 세포 표면을 나노필름으로 코팅하는 방법을 통해 세포에 전달되는 다양한 외부 자극들을 제어할 수 있음을 보여준다. 특히, 해당 연구는 나노필름의 구성 및 구조 제어를 통해 세포에 전달되는 자극을 정교하게 제어한다는 점에서 나노-바이오 융합 기술의 발전에 기여할 것으로 기대된다.

Research on controlling the physical and chemical properties and enhancing biological or pharmacological functions of biomaterials through surface modification has been continuously conducted and developed in diverse research fields. Among various methods of surface modification, multilayer nanofilms, a simple and biocompatible method, formed by physical adsorption of polymers can precisely control the surface. In addition, there are few limitations on the material constituting the nanofilm or the type of substrate, so that such nanofilms can be applied to various research field. In this study, experiments were conducted to enhance the function of biomaterials by controlling the structure and composition of these polymer-based nanofilms. In particular, data science, which is in the limelight as next-generation research paradigm, was introduced as a new method for understanding the properties of nanofilms that are changed by the materials and the experimental conditions. Through this study, the possibility of implementing an experimental data analysis platform that converts various numerical values generated in nanofilm experiments into data and estimates variable relationships through regression analysis was confirmed. The results are expected to contribute to improving the efficiency of various experiments based on polymer nanofilms. And a nanofilm was developed to control the release of growth factor, which is macromolecular protein. The amount of growth factor encapsulated in nanofilm was increased at the trilayer-structured multilayer nanofilm, and its release rate was controlled by blending layer in which two different polymers were mixed. It is expected that the efficiency of research can be increased depending on the cell type and culture purpose through the functional cell environment modified with such a nanofilm. In addition, functional extracellular matrices capable of controlling the differentiation of cells or enhancing cellular functions were implemented by coating a nanofilm on the cell membrane. A nanofilm composed of a polymer that generates chondrogenic differentiation was produced to improve the chondrogenesis efficiency of mesenchymal stem cells, and the extracellular matrix with increased stiffness was used to improve osteogenic differentiation efficiency of mesenchymal stem cells. Finally, by coating a flexible nanofilm containing a substance that inhibits apoptosis on the surface of induced pluripotent stem cell, it was possible to maintain its pluripotency and to promoting differentiation of embryoid body. Therefore, the results of this study show that various external stimuli delivered to cells can be controlled through the cell culture environment or the method of coating the cell surface with a nanofilm. In particular, this study is expected to contribute to the development of nano-biotechnology in that it precisely controls the stimulus delivered to cells through the control of the composition and structure of the nanofilm.
