In particular, small extracellular vesicles that are a subtype of cancer cell-derived extracellular vesicles can cause cancer progression and metastasis. These vesicles can provide information on several mechanisms, treatments, and diagnoses associated with cancer. This study has focused upon the isolation of these small extracellular vesicles derived from cancer cells. Although there are various methods for the separation of exosomes, they do have some limitations, and thus it is difficult to select the most efficient separation method. In this study, we used a small-scale tangential flow filtration (TFF) system for the separation of exosomes, and by adding a filter, and we developed a system that operates two filters in tandem to shorten the running time and increase the yield, purity, reproducibility, and scalability. The limitations of the existing separation method were improved. Furthermore, we used this system to identify the pattern of secretion of the small extracellular vesicles in cell culture media containing a minimum number of cells from various cancer cell lines and present the potential for drug screening by using drugs that inhibit the secretion of small extracellular vesicles. As a result, it has been shown that this tandem-based TFF system can be used as a tool for basic study and therapeutic research.
