Purpose: The purpose of this research is to identify single-nucleotide polymorphisms (SNPs) and their association with health related lifestyle on gout using genome data from Korean Genome and Epidemiology Study (KoGES).
Method: This research is a secondary analysis study that analyzes epidemiologic and genomic information with the utilization of HEXA based on metropolitan area and KARE based on the local community for KoGES. GWAS analysis was performed using K-chip through the Plink genome analysis program. A total of 18,927 people were analyzed, including HEXA's gout group of 438 people and the control group of 18,489 people for the discovery stage. A total of 3,063 people were analyzed, including KARE's gout group of 326 people and control group of 2,737 for the replication stage. And finally, a meta-analysis was performed using two cohorts. We analyzed the effects of lifestyle factors such as eating habits, physical activity, alcohol consumption, and smoking on gout. After identifying the association between GWAS-derived SNPs and health-related lifestyle factors, the PRS of SNPs was calculated to confirm the interaction between PRS and health-related lifestyle factors.
Result: 15 SNPs related to gout were found, among which rs1481012 of the ABCG2 gene (4:89039082_A/G) located on chromosome 4 was newly discovered (p=2.46E-11). When examining the interaction between SNP and health-related lifestyles, the rs3109823, located in the ABCG2 gene on chromosome 4, is associated with smoking. rs11936395, located in the SLC2A9 gene on chromosome 4, was significantly associated with average exercise amount per cycle. rs11066325 located in PTPN11 on chromosome 12, showed a significant association with the number of exercise per week, smoking, and drinking, respectively. It was also found that the rs20888548, located in the APP gene on chromosome 21, and the amount of Soju consumed per serving significantly interact with each other. In addition, it was verified that the interaction between PRS and duration of smoking affects gout.
Conclusion: This study found novel SNPs related to gout and identified their association with health-related lifestyle in Korean population. It provides basic data for predicting causative factors on gout and developing prevention and intervention programs in clinical and nursing perspective.
