Caveolin-1 (CAV1) is the main protein component of caveolae, which regulate various cancer-related processes such as tumor growth/survival, cancer migration/metastasis, and therapy resistance. In a previous study, NRF2 knockdown in colon cancer cell line HT29 facilitated cell motility in response to phorbol 12-myristate 13-acetate (PMA) treatment and showed differential gene expression patterns when compared to the control HT29. In this study, I investigated the role of CAV1, which is one of differentially regulated genes in NRF2 knockdown HT29 (shNRF2), in PMA-induced cancer motility/migration. The induction fold of CAV1 level in PMA-treated HT29 was higher in shNRF2 than that in PMA-treated control HT29. Silencing of CAV1 largely suppressed PMA-induced motility in shNRF2, which implies the critical role of CAV1 in PMA-mediated cell motility. In order to investigate the molecular event underlying PMA-induced CAV1 expression, I have examined PMA-associated signaling pathways such as extracellular signal-related kinase (ERK), Jun N-terminal kinase (JNK), and protein kinase B (AKT). Of note, ERK activation level was higher in PMA-treated shNRF2 cells, and the treatment of the ERK inhibitor U0126 suppressed cell motility and diminished CAV1 expression. As a cause of ERK activation, I observed the involvement of reactive oxygen species (ROS). The level of ROS in PMA-treated shNRF2 cells was higher than in the control cells, and the treatment of N-acetylcysteine, an inhibitor of ROS, significantly suppressed cell motility along with reduced CAV1 and p-ERK levels. Additionally, the potential direct regulation of CAV1 by NRF2 was notable. The basal level of CAV1 was diminished by NRF2 knockdown and conversely, genetic activation of NRF2 increased CAV1 expression. Taken together, these results indicate that enhanced motility of PMA-treated NRF2 knockdown colon cancer cells is mediated by ROS/ERK/CAV1 activation.
