BH4에서 대표적인 AHL-lactonase로 알려진 phosphotriesterase (PTE) family type인 QsdA 단백질과 α/β hydrolase type인 JydB 단백질을 암호화하고 유전자를 각각 결손 시킨 돌연변이 균주와 이 두 가지 유전자를 동시에 결손 시킨 이중 돌연변이 균주를 사용하여 Rhodococcus sp. BH4 야생형 균주와 P. carotovorum에 대한 QQ 활성을 조사하였다. 그 결과 Rhodococcus sp. BH4 야생형 균주와 qsdA 유전자가 돌연변이된 균주(ΔqsdA)에서는 AHL을 분해했고, jydB 유전자가 돌연변이된 균주(ΔjydB)와 qsdA, jydB 두 유전자가 돌연변이된 균주(ΔqsdAΔjydB) 에서는 AHL을 분해하지 못했다. 이러한 결과는 jydB 유전자가 좀더 AHL 분해에 관여하고 있다는 것을 시사한다. 또한 co-culture시 galacturonase의 활성은 Rhodococcus sp. The bacteria sense the concentration of the signal molecule and recognize it's concentration of surrounding cells. In many pathogenic bacteria, the AHL-based QS system regulates the expression of pathogenicity-related genes. Meanwhile the strategy of blocking the QS system to suppress the expression of pathogenicity-related genes is called quorum quenching (QQ). Therefore QQ could be potential biocontrol strategy against pathogenic bacteria. Pectobacterium carotovorum is a representative strain that causes plant soft rot diseases via AHL-based QS system. The production of biofilm, pectin lyase and galacturonase, which are typical virulence factor produced by P. carotovorum, are controlled by the AHL. In the mean time Rhodococcus strains have been reported to have prominent QQ activity, showing efficient AHL degradation. Therefore, it is interesting to examine the potential possibility that Rhodococcus strain, AHL degrader, could be used to develop biocontrol agent against P. carotovorum, AHL producer. In this study, we analyzed various quorum quenching Rhodococcus strains which were isolated in the laboratory. They have two types of AHL-lactonase, QsdA and JydB which belongs to phosphotriesterase family and α/β hydrolase superfamily, respectively. We examined QQ activity of Rhodococcus sp. BH4 wild and three AHL-lactonase mutants (ΔqsdA, ΔjydB, ΔqsdA ΔjydB) against P. carotovorum. Additionally, we compared QQ capability of wild and AHL-lactonase mutants to deduce the roles of QsdA and JydB in Rhodococcus sp. BH4. Results revealed that Rhodococcus sp. BH4 degraded efficiently AHLs which were produces by P. carotovorum when the two strains were cocultured. And analysis of the AHL-lactonase mutants suggesed that JydB has higher AHL-degrading activity than QsdA. In addition, the galactronase activity was decreased when P. carotovorum was cocultured with the wild-type strain compared to the mutant strains because of AHL degradation by Rhodococcus sp. BH4. In case of pectin lyase activity, when coculturing with P. carotovorum double mutant (ΔqsdA ΔjydB) showed high activity compared to wild type strain. In conclusion, these results suggest that AHL-degrader Rhodococcus has QQ acitity against AHL-producer P. carotovorum.
