The present study evaluated activities associated with the anti-fatigue in a hot water extract of the two fruits (Stauntonia hexaphylla (Thunb.) Decne. and Vaccinium bracteatum Thunb. complex extract, SVE). Stauntonia hexaphylla (Thunb.) Decne. was obtained from the cultivation area of Jangheung, Jeollanam-do, and the fruit of the Vaccinium bracteatum Thunb was obtained from the Goheung community, Jeollanam-do. Antioxidant evaluation was conducted to evaluate the activity of mature and immature fruits, and the final test sample was determined as the test material for the immature fruit for Stauntonia hexaphylla (Thunb.) Decne. and the mature fruit for the Vaccinium bracteatum Thunb. In the complex ratio evaluation, as a result of evaluating the indicators related to anti-inflammatory, the Stauntonia hexaphylla (Thunb.) Decne. and the Vaccinium bracteatum Thunb. extract were finalized as a 1:1 complex ratio. It was induced by oxidative stress in myotubes of the immortalized mouse myoblast cell line C2C12 using H2O2 in vitro, and produced exercise-induced fatigue in Institute of Cancer Research mice in vivo by exhaustive swimming. The findings in vitro revealed decreased viability and ATP contents of C2C12 myotubes with oxidative stress and that SVE (50, 100, and 200 μg/mL) significantly increased the cell viability and ATP contents. A significant decrease in reactive oxygen species (ROS) generation indicated that the ROS scavenging activity of SVE underlies the cytoprotective effect against H2O2-induced oxidative stress. The results in vivo showed that 50 (low dose) and 200 (high dose) mg/kg of SVE significantly increased the duration of time that the mice exhaustively swam compared to control mice. Low and high doses of SVE caused a significant decrease in serum lactate levels and lactose dehydrogenase (LDH) activity compared with the control, whereas serum nonesterified fatty acid (NEFA) levels were significantly increased only in the high-dose group. Intramuscular glycogen contents were significantly increased in the SVE group, compared with the control group. The mRNA expression of factors that regulate fatty acid beta-oxidation in the mitochondria was also increased in muscle tissues. When the levels of catalase (CAT), superoxide dismutase (SOD), glutathione-S-transferase (GST), and glutathione (GSH) were measured in the mice liver tissue, higher activities of the antioxidant enzymes and higher GSH content were noted across the SVE treatment groups than in the control mice. In addition, malondialdehyde (MDA) levels were significantly decreased in SVE compared with control mice, indicating that SVE prevented lipid peroxidation caused by exercise. These results suggested that SVE prevented the decrease in antioxidant activities during oxidative stress, and enhanced muscle functions via glycogen and fatty acid oxidation, thus improving recovery from exercise-induced fatigue.
