SDC-U1을 Polyvinyl alcohol(PVA)/Alginate 또는 Alginate비드에 고정 후 QS 신호분자인 Acyl homoserine lactone (AHL)의 분해를 확인한 결과 Alginate 비드에 고정시켰을 때에는 AHL 분해 활성이 충분히 나타났으며, PVA/Alginate 비드에 고정화 하였을 때는 제조과정에서 박테리아가 pH와 삼투압에 의한 영향을 받아 활성을 대부분 잃는 것으로 확인되었다. 제조된 QQ 비드가 MBR의 막오염을 저감할 수 있는지 확인하고자 Lab-scale MBR을 설치하여 산업폐수처리용 MBR에서 가져온 슬러지를 주입한 후 디스플레이 공정에 현상액으로 사용되는 Tetramethylammonium hydroxide (TMAH)를 함유한 실제 산업폐수를 유입시켜 연속 운전하였다. Phase 1에서 QQ 비드를 넣지 않은 대조군 MBR에 비해 QQ 비드를 넣었을 때, 막간차압이 30 kPa까지 도달하는데 걸린 운전 시간이 약 40% 증가하였다. 또한 Phase 2에서 미생물이 고정되지 않은 비드를 주입한 대조군 MBR에 비해 QQ 비드를 주입한 MBR은 운전시간이 약 50% 증가하여, QQ 비드에 의한 막오염의 지연이 비드의 물리적 세척효과에 의한 것이 아님을 확인하였다. 또한 오염된 분리막을 Live/Dead 염색한 후 공초점 레이저주사현미경을 통해 관찰한 결과 분리막의 오염에 있어 생물막의 형성이 주된 원인임을 확인하였다. 이 연구에서 TMAH 독성 물질을 처리하는 슬러지에서 분리된 Bacillus cereus SDC-U1을 Alginate 비드로 고정하였을 때 활성이 유지되며, 이를 MBR에 적용 하여 생물막오염을 저감할 수 있음을 확인하여, 산업폐수 처리용 MBR에서도 QQ 기술이 충분히 적용될 수 있음을 보였다. 이 결과는 앞으로 다양한 종류의 산업폐수를 처리하는 MBR에서 각각의 특성에 맞는 QQ 기술을 개발하는 연구가 활발히 진행 될 수 있는 밑거름이 될 것으로 예상된다.

Membrane biofouling in MBR can cause problems such as increased energy consumption and cleaning costs and shortening of membrane life. One of the methods to reduce biofouling is the quorum quenching (QQ) technology that interferes with quorum sensing (QS) based bacterial communication. Since microorganisms promote biofilm formation using the QS mechanism, interruption of QS in the MBR process may delay biofilm growth on the surface of the membrane. Recently, many studies have been conducted to reduce membrane biofouling in MBR for domestic wastewater treatment by applying QQ technology, but there are few studies on the effect of QQ on the MBR treating industrial wastewater containing toxic substances. Therefore, we tried to check whether the QQ technology can be applied to MBR treating industrial wastewater in this study. After entrapping Bacillus sp. SDC-U1, a QQ bacterium isolated from industrial wastewater, to polyvinyl alcohol (PVA)/Alginate or Alginate beads, the degradation of acyl homoserine lactone (AHL), a representative QS signal molecule, was analyzed. Alginate beads showed sufficiently QQ activity, whereas SDC-U1 lost their activity in PVA/Alginate beads due to the influence of pH and osmotic pressure during the manufacturing process. To check whether the QQ beads can reduce membrane biofouling in MBR, lab-scale MBR was continuously operated with inoculum sludge and influent wastewater from MBR plant that treats industrial wastewater containing toxic Tetramethylammonium hydroxide (TMAH), which is used as a developer in the display industry. In Phase 1, when QQ beads were added, the operating time required for the transmembrane pressure to reach 30 kPa was increased by about 40% compared to the control MBR without QQ beads. In Phase 2, the operation time of the MBR injected with QQ beads increased by about 50% compared to the control MBR injected with vacant beads, which confirmed that the delay in membrane biofouling by the QQ beads was not due to the physical washing effect of the beads. In addition, as a result of observation through a confocal laser scanning microscope after Live/Dead staining of the fouled membrane, it was confirmed that the biofilm formation was the main cause of the membrane fouling. In this study, it was confirmed that membrane biofouling can be reduced when SDC-U1 isolated from TMAH-containing sludge is entrapped in alginate beads and applied to MBR, which demonstrates that QQ technology can be successfully applied to MBR for industrial wastewater treatment. This result is expected to serve as a foundation for intensive research to develop QQ technology for targeting each MBR that treats specific type of industrial wastewater in the future.
