Although they have their own genomes, the majority of their proteins are encoded by nuclear genes, translated by cytosolic ribosomes, and imported via outer and inner membrane translocon complexes. However, it is not fully understood how the folding properties of these proteins affect their post-translational transport into these organelles. To address this issue, I examined the import behavior of chloroplast and mitochondrial preproteins fused to superfolder GFP (sfGFP), a more stable and better-folded than GFP and extra-superfolder GFP (esGFP), a more stable and better-folded than sfGFP to determine whether they would hinder the import process. Surprisingly, chloroplast preproteins fused to GFP and sfGFP and esGFP had comparable import efficiencies. Moreover, chloroplast preproteins fused to esGFP imported more efficiently than sfGFP and GFP, even though there was a mutation in critical functions in import process. By contrast, the fusion of mitochondrial preproteins to sfGFP and esGFP impeded their import into mitochondria. Based on these results, I propose that chloroplast import machinery is more tolerant of preproteins with different folding status.
