Although research on climate change and the related ecological risk for marine organisms is increasing globally, the mesocosm scale assessment of ecosystems remains limited. Therefore, in this study, I developed an adaptable indoor mesocosm system to study several aspects of global oceanic climate change and evaluate the physiological and ecological effects on seaweed species. Chapter 1 describes the development of an indoor mesocosm that can simulate climate change conditions. A device that could control gas concentrations to simulate various oceanic climate conditions that could appear in the future was developed, including ocean acidification, ocean warming, and deoxygenation. The environmental treatments of the laboratory mesocosm was established by injecting mixture gases into the seawater tank. CO2 concentrations of mixture gases were adjusted using a mass flow controller until target CO2 conditions were established. A deoxygenation environment was established by adding a large amount of N2 gas and a small amount of CO2 gas. To confirm whether experimental CO2 conditions were satisfied, it was briefly confirmed using a commercialized NDIR CO2 sensor, and the chemical properties of carbonate chemistry were precisely assessed using the carbon dioxide analysis method. The simulated conditions of ocean acidification, warming, and deoxygenation were well-controlled. Application of this system allowed for the physiological and ecological study of seaweed species, as described in Chapters 2 and 3. In Chapter 2, the physiological and ecological effect of ocean acidification and warming was investigated in the subtropical seaweed Caulerpa sertularioides. Although the genus Caulerpa is invasive worldwide, C. sertularioides has not been found in domestic seaweed habitats in South Korea. To evaluate the invasive potential of C. sertularioides, the individual and combined effects of ocean acidification (doubled CO2 level) and warming (5℃ warmer than present) on the ecophysiological performance of C. sertularioides was investigated. Ocean warming and elevated CO2 increased the electron transport efficiency and oxygen evolution, respectively. The growth of C. sertularioides increased with increases in CO2 concentration and temperature. In particular, a synergistic effect was observed in the combined treatment (greenhouse) compared to the individual treatments. These results suggest that C. sertularioides has the potential to serve as an invasive marine seaweed with increasing ocean temperature and acidity. Chapter 3 describes the investigation of the ecophysiological effects of ocean acidification and deoxygenation on the green-tide forming alga Ulva ohnoi. On Jeju Island, South Korea, the genus Ulva frequently causes green tides due to the active uptake of nutrients. Therefore, the bloom potential of U. ohnoi was investigated under ocean acidification (doubled CO2 level) and deoxygenation (approximately 4 mg O2 L-1) conditions, which occur frequently in polluted coastal environments. With increasing acidification and deoxygenation events, the photosynthetic efficiency (as measured using chlorophyll fluorescence) did not increase, while oxygenic photosynthesis decreased, compared to the present conditions; nevertheless, both the climate change factors positively affected growth of thallus area. In particular, deoxygenation conditions contributed to rapid growth, and the combined effect of ocean acidification and deoxygenation increased the chlorophyll contents. These results suggest that ocean acidification and deoxygenation may provide conditions for sustained growth of green tide-forming species. In this study, an efficient and low-cost indoor mesocosm that can effectively control various climate change conditions was developed. Results of the two seaweed experiments conducted in this study support the idea that this mesocosm is a suitable tool for studying climate change. In particular, this study provides important data for predicting the rise and fall of seaweed-related environmental issues in the future oceanic environments using two species of seaweeds that have large ecological impacts in Korea. Through future studies applying a longitudinal approach and including a large number of marine organisms, ecophysiological data on ecologically important species will be accumulated that may help in understanding the patterns of succession in domestic benthic communities in response to oceanic climate change.
