Genomic DNA (gDNA) 라이브러리와 비교하여 WGA DNA 라이브러리의 경우 input DNA 양 감소에 관계없이 on-target rate, on-SNP rate에서 보다 안정적인 결과를 확보할 수 있었고, genotype recovery rate가 최대 27배까지 상당히 증가함이 확인되었다. 나아가, duplicate rate 비교를 통해 미량 DNA를 대상으로 WGA를 수행하면 라이브러리 complexity가 개선되는 것을 유추해볼 수 있었다. 본 연구에서는 법과학 실무에서 어려움을 겪는 미량 DNA 분석 결과를 개선하는 대안으로 WGA 수행의 효용성을 보여주었다. 본 연구에서 구축한 방법은 현재 사용하고 있는 방법으로는 해결이 어려운 미량 DNA 분석에 적용하여 개인 신원확인 특정율을 높이고 수사 단서 획득에 기여할 것으로 사료된다. 나아가, 본 연구를 기반으로 얻어진 identity-informative SNP에 대한 NGS 결과는 추후 법과학 실무에서 다양하게 활용 가능한 대규모의 forensic SNP 패널로의 확장 전략을 수립하는데 유용하게 활용될 것으로 사료된다.

However, DNA extracted from crime scenes is frequently degraded or recovered in low quantities, limiting the scope of subsequent genetic analyses and leading to unreliable results. With the introduction of next generation sequencing (NGS) into forensic genetics, NGS analysis of single nucleotide polymorphisms (SNPs) that can be analyzed in a short length has been in progress. Since most SNP markers have two alleles and thus have relatively low discriminating power, so a large number of SNP markers should be analyzed simultaneously to obtain high discriminating power. In order to flexibly expand the panel to analyze multiple SNP markers, hybrid capture-based target enrichment methods are attracting attention in forensic genetics. Until now, preparation of NGS libraries for hybrid capture of SNPs on trace amounts of DNA has been mainly performed using low-input DNA library preparation kits. However, the existing method requires a certain amount of input DNA, and library complexity decreases as the amount of input DNA decreases. In this study, to overcome these limitations and increase analysis efficiency, the whole genome amplification (WGA), which generates sufficient amplified DNA even from a single cell, was proposed as an alternative. Accordingly, we developed a procedure for hybrid capture-based MPS of 136 identityinformative SNPs for serially diluted samples to sub-nanogram levels and compared MPS results of the SNPs between genomic DNA (gDNA) and WGA DNA depending on the amount of input DNA. On-target rate, OnSNP rate, duplicate rate, and genotype recovery rate of target SNPs were investigated. In the case of the WGA DNA library, more stable results were obtained at the on-target rate and on-SNP rate regardless of the decrease in the amount of input DNA, and it was confirmed that the genotype recovery rate was significantly increased up to 27 times compared to the gDNA library. Furthermore, it was possible to infer that library complexity was improved when WGA was performed on trace DNA through comparison of duplicate rates. According to the results of this study, the utility of WGA application was demonstrated in hybrid capture-based NGS analysis targeting trace amounts of DNA. The results of this study suggest that performing WGA can increase the efficiency of hybrid capture-based NGS for low quantities of DNA, a longstanding challenge in forensic practice. Furthermore, the NGS data on identity-informative SNPs obtained through this study will be useful in establishing an expansion strategy to a large-scale forensic SNP panel that can be used in various ways in forensic practice.
