종양 미세 환경 (Tumor microenvironment, TME) 반응성 나노 플랫폼을 통한 표적 종양 치료는 정상 세포에는 최대한 영향을 주지 않고 암세포만을 선택적으로 죽이기 위해 사용되는 치료 전략이다. 대표적인 항암 치료법 중 하나인 화학역학요법 (Chemodynamic therapy, CDT)은 암세포 내부의 과산화수소를 금속 이온의 촉매 작용 하에 독성이 강한 하이드록실 라디칼로 전환시켜 암세포의 성장을 억제하고 결국에는 사멸에까지 이르게 하는 새로운 전략인데, 레이저, 전기 또는 초음파와 같은 외부 에너지의 개입이 없어도 종양 미세 환경에 풍부하게 존재하는 내인성 물질 (예: 포도당, 과산화수소)에 의해 화학역학요법의 촉매 반응이 시작된다는 장점이 있다. 화학역학요법을 위한 치료제로서, 우수한 효소 모방 활성을 가지는 나노물질, 즉 나노자임이 다양한 나노치료적 접근법에 널리 적용되어 왔다. 초미립자 크기, 열적 안정성, 높은 촉매 활성, 간단한 합성 과정, 우수한 내구성이라는 장점을 지닌 나노자임을 약물 운반체 역할을 하는 나노 구조체에 포집하여 치료제로서의 생체 적합성을 높이는 연구 또한 꾸준히 이루어지고 있다. 그러나 화학역학요법에는 단점 또한 존재하는데, 불충분한 양의 암세포 내인성 과산화수소와 강력한 항산화제가 하이드록실 라디칼의 생성을 억제하여 치료 효과를 감소시킬 수 있다는 것이며, 이러한 한계를 극복하기 위해서는 화학역학요법을 다른 치료법과 결합시켜 해당 단점을 보완하고 단일요법보다 높은 치료 효율과 낮은 부작용을 보이는 복합적 방식의 치료법 개발이 요구된다. 나아가, 나노자임 기반 나노치료제의 생체 적합성, 치료적 효율성 및 항암제로의 응용 가능성을 확인하기 위해서는 목적에 맞는 생체 외 (in vitro) 임상적 분석이 반드시 수행되어야 한다. 하지만, 흡광도 측정에 기반한 기존의 생체 외 세포 분석법으로는 유색의 항암 나노자임 또는 나노복합체의 효능을 검증하기 어려우며, 이로 인해 간혹 거짓 양성/음성과 같은 분석 오류를 범할 수 있다는 결정적인 단점이 있다. 따라서 나노물질 기반 항암제의 새로운 발견을 위해서는 기존 분석법의 대체가 가능하고 유색 약물의 효능 평가가 쉽고 빠르며 간편하게 이루어지는 분석 플랫폼의 개발이 필요하다. 세포 기반 칩은 살아있는 세포를 배양하기 위한 소형화된 플랫폼으로, 암세포 증식을 억제하고 사멸을 유도하는 항암제의 치료 효과를 모니터링하는 데 유용한 장치가 될 수 있다. 세포칩의 장점으로는 조작이 용이하고 신속 및 정확하며 민감한 분석이 가능하다는 점을 들 수 있는데, 이를 전기화학적 센싱 기법과 접목시켜 개발한 것이 바로 전기화학적 세포칩이다. 분석 절차가 간단하고 특이적인 표지가 필요 없으며, 비침습적인 방식의 세포 센싱 플랫폼을 구현할 수 있어서 항암 약물의 생체 외 효능을 평가하기 위한 장치로써 널리 연구되어 오고 있다. 본 학위 논문의 2장에서는, 전기화학적 방식을 이용한 세포 패터닝 기법과 세포칩의 개발 및 항암 약물 스크리닝 플랫폼으로의 응용이 소개되었다. 불소 도핑 주석 산화물 (Fluorine-doped tin oxide, FTO) 전극 표면에서 발생하는 전기장이 암세포 (Skin malignant melanoma, SK-MEL28) 내/외부 이온 불균형을 유발하여 세포가 손상을 입고 사멸하는 방식을 통하여, 전기적 자극을 받지 않는 부분의 모양대로 세포 패턴을 구현하였다. 전해질, 인가 전압의 세기 및 시간과 같은 전기화학적 조건의 차이를 이용하여 세포 패터닝 기법을 개발하였고 이를 다양한 종류의 세포에 적용하였다. 또, 전기화학적으로 세포가 패턴된 기판을 고분자 필름 기반의 미세 유체 칩과 결합시켜 항암 약물 (사포닌)의 주입에 따른 생존 세포 수 변화를 정성적/정량적으로 분석하는 방식으로 약물 스크리닝을 실시하였다. 3장에서는, 세포 기반의 전기화학적 바이오센서의 개발 및 이를 이용한 암세포 감지와 항암 약물의 효능 평가법이 소개되었다. 전기화학적으로 세포 감지가 가능한 기법/플랫폼을 개발하여 항암 약물 (사포닌)이 SK-MEL28암세포에 미치는 성장 억제 및 사멸 효과를 민감하고 빠르게 평가하였다. 전기촉매적 특성과 생체적합성 향상을 위하여 금 나노입자 (Gold nanoparticles, AuNPs)와 폴리라이신 (Poly-L-lysine, PLL) 분자를 투명 FTO전극 위에 퇴적 및 코팅하였다. 일정 전압을 세포가 자라는 전극에 인가하면 표면에 고정된 세포와 전극 표면 간의 직접적인 전자 전달로 인해 전기적 신호가 나타나게 되고, 결국 측정 전류값이 세포 수에 정비례함을 이용하여 약물의 항암 효능이 암세포의 생존율에 미치는 영향을 전기화학적으로 정량화하였다. 전기화학적 측정에서, 암세포의 선형 감지(검출) 범위는 장치 당 2,880 – 40,000 개로 나타났으며, 해당 범위에서 세포 수와 전류 세기의 사이의 선형성을 확인하였다 ("R" ^"2" = 0.9952). 또한, SK-MEL28 암세포에 대한 사포닌의 항암 효과가 20 "μM" 이상의 농도에서 명확하게 나타났으며, 이는 기존 분석법과 매우 일치하는 결과임을 확인하였다. 결과적으로, 유색 약물의 분석을 할 때에 측정 파장이 겹친다는 단점이 있던 비색 측정법보다 더 신속하고 (< 2분) 민감하며 (검출한계: 2,880 세포/장치) 비침습적인 방식의 항암 효능 평가법을 개발하였다. 4장에서는, 종양 미세환경에 반응하는 항암 나노복합체의 개발 및 항암제로서의 치료 효능 검증에 대한 연구가 소개되었다. 과산화효소 활성을 가지는 나노자임 (Prussian Blue nanoparticle. Targeted tumor therapy through tumor microenvironment (TME)-responsive nanoplatforms is an emerging treatment strategy used to selectively kill cancer cells. Among the representative therapeutics, chemodynamic therapy (CDT) is a novel strategy that converts endogenous hydrogen peroxide (H2O2) in cancer cells into highly toxic hydroxyl radicals (∙OH) under the catalysis of metal ions. Even without external energy input, such as laser, electricity, or ultrasound, the catalytic reaction in CDT can be initiated by the glucose abundant in the TME, thus promoting ROS generation to inhibit the tumor cell growth. As a CDT agent, nanomaterial-based artificial enzymes, called nanozymes, have been widely used in many nanotherapeutic approaches based on their excellent enzyme-mimicking activity. Under the advantages of ultrafine particle size, thermal stability, high catalytic activity, easy preparation, and good durability, they can be incorporated into various nanostructures that function as drug carriers to enhance biocompatibility. In CDT, however, the insufficient intratumoral H2O2 level and robust antioxidants in cancer cells might suppress the production of ∙OH, thereby reducing the therapeutic effects. To overcome this limitation, it is required to reinforce CDT by combining it with other therapeutic modalities, yielding the multimodal therapies with higher efficiency and lower systemic side effects than the monotherapy. Furthermore, clinical evaluation of the nanozyme-based nanotherapeutic drugs for cancer therapies must be conducted to determine the efficiency and biocompatibility for their therapeutic usage. However, traditional assays based on absorbance measurements have a critical drawback that it is difficult to verify the efficacy of anticancer nanocomposite having its unique color. Therefore, it is required to develop simple and accurate methods that can be implemented easily for discovering new anticancer agents. A cell-based chip is a miniaturized platform for culturing living cells and can be a useful device for monitoring the therapeutic effects of anticancer drugs that inhibit cancer cell proliferation. Due to the ease of operation, rapid, accurate, and sensitive analysis, it has been applied in combination with an electrochemical sensing technique for in vitro cell culture analysis. The electrochemical cell chips have been widely used as simple, label-free, and non-invasive techniques for the evaluation of the effects of anticancer drugs on cancer cells. In chapter 2 of this dissertation, a simple electrochemical approach for arranging cells into various patterns was introduced to enable rapid and accurate localization for cell chip formation. A voltage pulse was applied directly to the pre-patterned conducting substrate, causing cellular damage, and rapidly forming live/dead cell arrays. The array platform, which focuses on assay miniaturization, was applicable to cell-based drug screening by determining the anticancer effect of drugs on various types of cells. In chapter 3, a novel and simple cell-based electrochemical biosensor, referred to as a cytosensor, was proposed to investigate the electrochemical behavior of human skin malignant melanoma (SK-MEL28) cells and the anticancer effect of saponin on cell viability. To enhance both electrocatalytic properties and biocompatibility, gold nanoparticles were electrochemically deposited onto a conductive substrate, and poly-L-lysine was further added to the electrode surface. Electric signals from SK-MEL28 cells on the electrodes were obtained from cyclic voltammetry and differential pulse voltammetry. The cathodic peak current was proportional to the cell viability and showed a detection range of 2,880 – 40,000 cells per device with an excellent linear cell number-intensity relationship (R2 = 0.9952). Furthermore, the anticancer effect of saponin on SK-MEL28 cells was clearly established at concentrations higher than 20 μM, which was highly consistent with conventional assays. Moreover, the developed electrochemical cytosensor for evaluating anticancer effects enabled rapid (< 2 min), sensitive (LOQ: 2,880 cells/device), and non-invasive measurements, thus providing a new avenue for assessing the anticancer drugs in vitro. In chapter 4, we developed a nanosized zeolitic imidazolate framework-8 (ZIF-8) that simultaneously contains natural glucose oxidase (GOx) and Prussian blue nanoparticles (PBNPs), which act as a peroxidase-mimicking nanozyme, to construct TME-activatable nanocomposites (denoted as ZIF@GOx@PBNPs) for effective tumor suppression. In weak acidic TME, GOx effectively catalyzed the oxidation of intratumoral glucose to H2O2 and gluconic acid. Meanwhile, PBNPs released from the ZIF-8 framework dissociated by acidic pH convert the generated H2O2 into harmful ∙OH radicals. In this way, the cascade catalytic reactions of ZIF@GOx@PBNPs enhanced reactive oxygen species production and caused intracellular oxidative damage to tumor cells, resulting in remarkable inhibition of tumor growth. Given the significant antitumor efficiency both in vitro and in vivo, ZIF@GOx@PBNPs could be applied as a promising therapeutic platform enabling starvation/chemodynamic synergism, high therapeutic efficiency, and minimal side effects. These results above clearly demonstrate the potential of nanozyme-integrated MOF nanocomposites as tumor-targeted synergistic therapeutics for effective cancer treatment. Furthermore, the development of electrochemical cell chips and its application for in vitro analysis can be extended for the assessment of other anticancer drugs in a rapid and sensitive way.

Moreover, the developed electrochemical cytosensor for evaluating anticancer effects enabled rapid (< 2 min), sensitive (LOQ: 2,880 cells/device), and non-invasive measurements, thus providing a new avenue for assessing the anticancer drugs in vitro. In chapter 4, we developed a nanosized zeolitic imidazolate framework-8 (ZIF-8) that simultaneously contains natural glucose oxidase (GOx) and Prussian blue nanoparticles (PBNPs), which act as a peroxidase-mimicking nanozyme, to construct TME-activatable nanocomposites (denoted as ZIF@GOx@PBNPs) for effective tumor suppression. In weak acidic TME, GOx effectively catalyzed the oxidation of intratumoral glucose to H2O2 and gluconic acid. Meanwhile, PBNPs released from the ZIF-8 framework dissociated by acidic pH convert the generated H2O2 into harmful ∙OH radicals. In this way, the cascade catalytic reactions of ZIF@GOx@PBNPs enhanced reactive oxygen species production and caused intracellular oxidative damage to tumor cells, resulting in remarkable inhibition of tumor growth. Given the significant antitumor efficiency both in vitro and in vivo, ZIF@GOx@PBNPs could be applied as a promising therapeutic platform enabling starvation/chemodynamic synergism, high therapeutic efficiency, and minimal side effects. These results above clearly demonstrate the potential of nanozyme-integrated MOF nanocomposites as tumor-targeted synergistic therapeutics for effective cancer treatment. Furthermore, the development of electrochemical cell chips and its application for in vitro analysis can be extended for the assessment of other anticancer drugs in a rapid and sensitive way.
