Plastic, a synthetic polymer, has been widely used due to various advantages such as low cost, light weight, and durability. Currently, solutions are being requested to solve the environmental pollution caused by indiscriminate use of plastic. In this study, we selected novel plastic-degrading microorganisms, and confirmed chemical changes of plastic reacted with microorganisms by FT-IR analysis to explore plastic biodegradation mechanisms. In addition, a culture solution in which plastic and microorganisms were cultured was analyzed by GC-MS to identify intermediates that may be generated during the plastic biodegradation process. Based on these results, alkane hydroxylase (AlkB), an enzyme that has the potential to act as the first enzyme in plastic biodegradation mechanisms, was selected, and the hydroxylation activity of AlkB on plastic was confirmed through experiments such as enzyme reaction and gene transcriptional analysis.
