Methacrylate groups in MeHA were cross-linked to each other by photo-initiator and UV irradiation to produce a hydrogel microneedle. The characteristics of hydrogel microneedles prepared with MeHA and HA/MeHA mixture were evaluated to establish an optimal microneedle system for ISF extraction. The mechanical strength of the microneedles was assessed by a texture analyzer to demonstrate skin permeability. Furthermore, the swelling property was evaluated by measuring the extracted mass of the buffer during a specific time. An artificial skin, agarose gel covered with parafilm, was produced to assess the permeability-swellability of the microneedle system assisted by ultrasound. Ultrasound was applied to the skin during the ISF extraction to increase the volume further. As a result, both the HA MeHA and MeHA microneedles showed sufficient skin permeability and ISF extraction property. HA MeHA microneedles showed better skin permeability and swelling ability than MeHA microneedles. Moreover, the ultrasound applied microneedle system to the skin enhanced the ISF extraction volume to 2-fold compared to the previous. Therefore, the HA MeHA microneedle system assisted by ultrasound is promising to contribute to developing a simple, cost-effective and patient-friendly diagnostic system alternative to the conventional invasive procedure.
