An enzymatic biofuel cell (EBFC) is a device that converts into electrical energy using chemical reactions that occur through the metabolic process of enzyme catalysts. In the case of EBFC, unlike general fuel cells, the mixed potential is not generated even when fuel is mixed due to the substrate selectivity of the enzyme used as a catalyst, so it is possible to construct a system without a separator (membrane). Since the composition is simple, the interaction between the enzyme used as a catalyst and the electrode is a factor that determines the overall performance. There are several factors that determine the performance of EBFCs, such as fuel delivery, enzyme catalyst stability, and electron transfer rate. In this study, biocompatible mediators that help enzyme catalysts transfer electrons and a method for increasing the electron transfer rate were developed. Glucose oxidase (GOX) was used for the anode and bilirubin oxidase (BOD) was used for the cathode as enzyme catalysts for the EBFC. GOX, the enzyme used in the anode, needs mediators to help electron transfer improve its performance because the active site is deep in the thick protein shell. Commonly used mediators are often toxic, so there is a limit to the application of implantable devices, so polydopamine (PDA), a biocompatible material, was used as a mediator to evaluate performance. When using carbon nanotube (CNT)/PDA/GOX/PEI as the anode and CNT/BOD as the cathode, the performance of open circuit voltage (OCV) and maximum power density (MPD) was 0.571 ± 0.007 V and 135.2 ± 5.45 μW cm-2, respectively, under oxygen saturation conditions in which 10 mM glucose was injected as fuel. BOD, an enzyme used in the cathode, has active sites very close to the outside of the protein shell, enabling direct electron transfer (DET) with the electrode. The active site of BOD, the T1 site, is positively charged under neutral pH conditions. Under neutral pH conditions, 4-Aminophthalic acid (4-APA), which gives off hydrogen ions and has two negatively charged carboxyl groups per molecule, is electrografted onto the surface of the electrode to form negative charges and improve the electron transfer performance of BOD. When using Buckypaper (BP)/4-APA/BOD as the cathode and BP/Tetrathiafulvalene (TTF)/GOX/Gelatin-glutaraldehyde (GA) as the anode, OCV and MPD showed 0.617 ± 0.004 V and 284.6 ± 1.67 μW cm-2, respectively, under air saturation conditions in which 10 mM glucose was injected as fuel.
